A method for non-surgical neutering or castration of a non-human mammal for AAV-mediated delivery of an anti-GnRH polypeptide to a non-human animal is described. More particularly, the animal is administered an adeno-associated virus (AAV) vector having an AAV capsid having packaged therein nucleic acid sequences comprising an AAV 5′ inverted terminal repeat (ITR), a sequence encoding a polypeptide which specifically binds gonadotropin releasing hormone (GnRH) under control of regulatory sequences which direct expression of the polypeptide, and an AAV 3′ ITR. A composition comprising the AAV-anti-GnRH may also be used for inhibiting tumor growth in a mammal with a cancer responsive to gonadal steroid hormones.

A prophylactic regimen for passively preventing infection with a pathogen which has a typical route of infection through the nasopharynx region of a subject, e.g., an airborne virus typically transmitted through coughing or sneezing. The method involves specifically targeting a subject's nasopharynx with a viral vector comprising an AAV capsid and carrying a nucleic acid sequence encoding an anti-viral neutralizing antibody construct operably linked to expression control sequences, in order to provide for high levels of expression of the anti-viral neutralizing antibody construct in the nasal airway cells. Optionally, the neutralizing antibody construct is expressed under a promoter which is regulated or induced by a small molecule which is delivered separately from the viral vector. In one embodiment, the method permits transfection of a subject's nasopharynx even where the subject has circulating neutralizing antibodies against the AAV capsid.

A method for tolerizing a mammalian subject to administration of a live virus carrying a gene for delivery to a cell of the subject is disclosed. The method entails administering to the subject a suitable amount of an inactivated virus prior to administration of the live virus. The prior administration of the inactivated virus suppresses anti-virus cytotoxic T cells, permitting longer transgene persistence once the live virus is administered, and permitting effective readministration of live virus.

A method for producing recombinant adeno-associated virus in the absence of contaminating helper virus or wild-type virus involves culturing a mammalian host cell containing a transgene flanked by adeno-associated virus (AAV) inverse terminal repeats and under the control of regulatory sequences directing expression thereof, an AAv rep sequence and an AAV cap sequence under the control of regulatory sequences directing expression thereof; and the minimum adenovirus DNA required to express an Ela gene product, an E1b gene product and an E2a gene product, and isolating therefrom a recombinant AAV which expresses the transgene in the absence of contaminating helper virus or wildtype AAV. This method obviates a subsequent purification step to purify rAAV from contaminating virus. Also provided are various embodiments of the host cell.

A recombinant adenovirus and a method for producing the virus are provided which utilize a recombinant shuttle vector comprising adenovirus DNA sequence for the 5' and 3' cis-elements necessary for replication and virion encapsidation in the absence of sequence encoding viral genes and a selected minigene linked thereto, and a helper adenovirus comprising sufficient adenovirus gene sequences necessary for a productive viral infection. Desirably the helper gene is crippled by modifications to its 5' packaging sequences, which facilitates purification of the viral particle from the helper virus.
