A composite biological-molecule-accretion material (10) is described for extracting desired target biological molecules from aqueous solution (24) and substantially permanently binding to those molecules. The material (10) includes a first component (10a &cir& _) designed with a binding affinity for proteins and peptides in such solution, and a second component (10b &cir& _) designed with a binding affinity for nucleic acids in such solution. The material (10) is capable of being formed in a body (12) for passing such solution through it, and such passing has the effect of removing the proteins, peptides and nucleic acids from such solution and binding them to the body. That binding is substantially permanent so that subsequent passage of additional aqueous solution will not unbind substantially the accreted proteins, peptides and nucleic acids. The first component (10a &cir& _) may include dextran-coated (10a &cir& _2) activated charcoal particles (10a &cir& _1), and the second component may include an anion-exchange resin and a cation-exchange resin. Preferred relative volumes of both components are 1:1. Also described is a method for extracting desired target biological molecules from aqueous solution and substantially permanently binding them to a solid, and a method of filtering target biological molecules out of water.

Disclosed are novel proteins, referred to as megakaryocyte growth and development factors (MGDFs; also generally referred to as Mpl ligands), that have a biological activity of stimulating the growth of megakaryocytes and augmenting the differentiation or maturation of megakaryocytes, ultimately to result in the production of platelets. MGDF derivatives comprising MGDF molecules attached to water soluble polymers, such as polyethylene glycol, are also disclosed, along with methods for their preparation. Also disclosed are processes for obtaining the MGDFs in homogeneous form from natural sources and producing them by recombinant genetic engineering techniques from mammals, including humans.

The chemical specie desirably is free of latent reactive groups that are activated upon activation of the latent reactive groups are simultaneously activated to cause formation via covalent bonding of a three-dimensional molecular network in which molecules of the chemical specie are covalently bonded to molecules of the coupling compound, and molecules of the coupling compound are covalently bonded to each other. Preferably, the three-dimensional matrix is formed as a coating upon a surface to which covalent bonds are formed upon activation of the latent reactive groups of the coupling compound.

The invention provides chemical methods for derivatizing natural and recombinantly-derived proteins or polypeptides containing cysteine residues, either naturally or through site specific mutageneses. The pharmaceutical compositions containing said derivatized proteins and/or polypeptides are formulated to provide stable, long-acting compositions of such proteins and/or polypeptides, previously difficult to achieve.
