    摘要: Apparatus and method for the separation of immiscible liquids are provided, and comprise the use of separator means having a surface which is preferentially "wettable" by one of the liquids to the substantial exclusion of another of the liquids. In use, the liquids are flowed from a common liquid source onto said surface with the one liquid being sorbed thereby to the substantial exclusion of the other liquid to thus substantially separate the liquids. The separator means surface may be maintained essentially level to retain the substantially separated liquid thereon, or may be inclined to result in the flow of the same therefrom under the influence of the force of gravity. Alternatively, the separator means may be made of a liquid-permeable material to permit the flow of the substantially separated liquid therethrough and therefrom under the influence of the force of gravity. The apparatus and method are particularly adapted to the substantial separation of aqueous sample liquids from hydrophobic isolation liquids which encapsulate the same; and are readily combinable with a sample liquid reaction device to effectively combine the sample liquid separation and reaction operations. The material of the separator means may be inherently hydrophobic, or may be rendered super-hydrophobic by the surface coating thereof with a super-hydrophobic coating material.

    摘要: A one-step method (and sphering agent/fixing agent useful therein) for treating mammalian red blood cells in a sample which can be effectively measured electrooptically for determination of red blood cell volumes, which comprises treating an anticoagulated whole blood sample with a single aqueous sphering agent/fixing agent reagent at a temperature in the range of 4°C to 37°C over a reaction period of up to 180 seconds employing a whole blood dilution factor of from 1:300 to 1:4,000 wherein said ona step, single reagent treatment provides isovoluments sphering of the erythrocytes in the semple independent of the concentration of said erythrocytes in the whole Diood sample and without inducing lysis of said erychwocytes in the resulting diluted semple. The preferred sptering agent is an alkali metal salt of an alkyl sulfate irr an amount 0.5 to 3.0 mg %, and the preferred fixing agent is an aldehyde in an amount from 0.05 to 0.3%.

    摘要: Dual liquid dispenser package is provided and is operable to supply liquids from liquid supply chambers to liquid reservoirs through liquid supply channels by centrifugal force-induced liquid displacement and flow attendant dispenser package rotation. The supply channels include means to greatly inhibit, if not totally prevent, liquid foaming attendant liquid flow from the supply chambers to reservoirs. Vents are included to vent the liquid supply chambers to atmosphere to prevent vacuum build-up therein, and include means preventing spillage of the liquids from the dispenser package through the vents. The dispenser package is particularly adapted for use in the supply of two reagent liquids to automated sample liquid analysis systems.

    摘要: A reagent for a specific binding assay, such as an immunoassay, is prepared by reacting both a partner of a specific binding pair and at least one hydrophilic substance, with haloalkyl moieties associated with the surface of a solid phase, and then recovering the reagent so prepared for any unreacted binding partner. Preferably, the alpha-haloalkyl is chloromethyl and the at least one hydrophilic substance comprises at least one protein, such as aibumin, and at least one non-proteinaceous amine to which the solid phase is impermeable, such as tris (hydroxymethyl) aminomethane. The specific binding assay reagent of the invention can be combined with other reagents to provide an assay composition.

    摘要: Lipid vesicles are formed by providing a lipidcoating on a deformable surface and fragmenting the lipid coating during hydration by deformation of the surface. The resulting fragments of the lipid coating are dispersed in the hydrating medium, so as to be subjected to multidirectional hydration, whereby formation of vesicles is accelerated. In a preferred embodiment, the surface is defined by a plurality of water-swellable particles formed of a polymeric material.

    摘要: The volume and/or index of refraction of a particle are determined by passing the particle through a beam of light directed along an optical path to produce at least one forward light scattering pattern;
    detecting and measuring the intensity of light at least within a selected portion of said at least one scattering pattern to generate at least a first signal and a second signal; and
    determining the volume and index of refraction of said particle from the magnitudes of said first and second signals. Apparatus for this purpose comprises means (26) for directing a light beam along an optical path,
    means (16) for passing said particle through said light beam to develop at least one forward light scattering pattern,
    means (42, 48) for detecting and measuring the intensity of light at least within a selected portion of said at least one scattering pattern and for generating a pair of first and second signals, and
    means responsive to said detecting and measuring means (42, 48) for determining and reporting the volume and/or the index of refraction of said particle from the magnitudes of said first and second signals. The method and apparatus are particularly useful for red blood cell analyses.
