A process to produce an aqueous solution of carbohydrates that contains C6-sugar-containing oligomers, C6 sugar monomers, C5-sugar-containing oligomers, C5 sugar monomers, or any combination thereof is presented. The process includes the steps of reacting biomass or a biomass-derived reactant with a solvent system including a lactone and water, and an acid catalyst. The reaction yields a product mixture containing water-soluble C6-sugar-containing oligomers, C6-sugar monomers, C5-sugar-containing oligomers, C5-sugar monomers, or any combination thereof. A solute is added to the product mixture to cause partitioning of the product mixture into an aqueous layer containing the carbohydrates and a substantially immiscible organic layer containing the lactone.

The present invention relates to the field of biology, in particular to a sugar preparation method by using biomass sweet potato dregs for microbial fermentation as a sugar source. The liquid sugar product prepared with the method of the present invention essentially comprises the ingredient of glucose. The method of the present invention is of simple process, high specificity, good product quality and high yield, and solves the serious environmental pollution problem of the sweet potato dregs, thus having a good industrial application prospect.

A fungal alpha-amylase is provided from Aspergillus fumigatus (AfAmy1). AfAmy1 has an optimal pH of 3.5 and is operable at 30-75 degrees C., allowing the enzyme to be used in combination with a glucoamylase and a pullulanase in a saccharification reaction. This obviates the necessity of running a saccharification reaction as a batch process, where the pH and temperature must be readjusted for optimal use of the alpha-amylase or glucoamylase. AfAmy1 also catalyzes the saccharification of starch substrates to an oligosaccharide composition significantly enriched in DP2 and (DP1+DP2) compared to the products of saccharification catalyzed by an alpha-amylase from Aspergillus kawachii. This facilitates the utilization of the oligosaccharide composition by a fermenting organism in a simultaneous saccharification and fermentation process, for example.

Fungal glucoamylases from Aspergillus fumigatus that are expressed in Trichoderma reesei host cells (AfGATR) are provided. AfGATRs, including AfGA1TR and AfGA2TR, exhibit high activity at elevated temperatures and at low pH, so AfGATRs can be used efficiently in a simultaneous liquefaction and saccharification process in the presence of alpha amylase, such as A spergillus kawachii alpha-amylase (AkAA). This greatly reduces the combined run time of liquefaction and saccharification reaction, where the pH and temperature must be readjusted for optimal use of the alpha-amylase or glucoamylase.

The present teachings provide a method for making a high glucose syrup at a low temperature. In some embodiments, the syrup contains reduced reversion reaction products. The method comprises contacting a starch substrate at a temperature below the starch gelatinization temperature with an enzyme blend comprising a high dose of alpha-amylase and a low dose of glucoamylase. In some embodiments, a blend of two glucoamylases is employed. In some embodiments, a debranching enzyme such as a pullulanase is employed. In some embodiments, the enzymes are staged by adding at different times, or at different temperatures. The present teachings provide for high glucose syrups with fewer reversion products, and allow for higher starch solubilization.

The present invention provides processes and systems for treating biomass and, e.g., making biofuels, such as bioethanol, from the biomass. More particularly, one process according to the present invention includes (a) inducing at least a first portion of a composition containing biomass and a working fluid to flow into a passage of a fluid processing apparatus, (b) injecting a high velocity transport fluid into the composition through a nozzle communicating with the passage of the fluid processing apparatus, whereby the transport fluid applies a shear force to the composition such that the working fluid is atomised and a vapour and droplet flow regime is formed downstream of the nozzle, (c) condensing the vapour and droplet flow regime, (d) transferring the composition to a first holding vessel, and (e) holding the composition in the first holding vessel at a first predetermined temperature for a first predetermined period of time, wherein a liquefaction enzyme is added to the composition prior to or during the process. Thereafter, the composition may be further processed to form a biofuel, such as, e.g., bioethanol.
