    摘要: Compositions and methods for the efficient transformation and regeneration of monocot plants are provided. The methods of transformation involve infection with Agrobacterium. In this manner, any gene of interest can be introduced into the monocot plant with high transformation efficiency and in low copy number. Transformed and regenerated monocot cells, tissues, plants, and seed are also provided. The invention encompasses regenerating transformed plants, transgenic seeds produced therefrom, and transgenic plants and transgenic seeds from subsequent generations.

    摘要: Compositions and methods are provided for the efficient transformation of a dicot plant. More particularly, compositions and methods of the present invention find use in agriculture for Agrobacterium-mediated transformation of a dicotyledonous plant. The compositions include cultivation media comprising high concentrations of sucrose and glucose. The cultivation media find use in methods directed to Agrobacterium-mediated transformation of a dicot plant with a gene of interest. In this manner, any gene of interest can be introduced into a dicot plant with high transformation efficiency and reduced tissue necrosis.

The invention includes a method of identifying gene expression modulating elements and gene insulator elements through use of the following steps: a) locating intergenic regions of a plant genome that are flanked by a gene on each side that have differing gene expressions b) taking that intergenic region or a portion of that intergenic region and adding it to a cassette comprising an isolated gene c) introducing the cassette into a plant cell d) analyzing expression of the isolated gene.
