One aspect of the invention is oligosaccharide-based therapeutics that interact with toxins and methods of uses thereof. In one embodiment the oligosaccharide-based therapeutics of the invention comprise polymeric particles with attached oligosaccharide binding moieties. The compositions of the invention can be used in the treatment of toxin-mediated diseases such as antibiotic-associated diarrhea and pseudomembranous colitis, including Clostridium difficile associated diarrhea.

In some cases, the anion-binding polymers have a pore volume distribution such that a fraction of the polymer is not available for non-interacting solutes above a certain percentage of the MW of the target ion for the polymer. In some cases, the anion-binding polymers are characterized by low ion-binding interference, where the interference is measured in, for example, a gastrointestinal simulant, relative to non-interfering buffer. Control agents that have a nitrogen-nitrogen bond covalently bonded to a thiocarbonyl moiety are provided for living-type free radical polymerization of a wide variety of monomers. These control agents provide superior properties for control of the polymerization and/or the properties of the polymers obtained and/or the monomers that may be polymerized. In some embodiments, a bulky group is pendant off the activated thiocarbonyl portion of the control agents. Multifunctional control agents provide the opportunity for a variety of structurally unique polymers, including block copolymers, stars and hyper-branched polymers.
