摘要: Provided herein are recombinant microorganisms having two or more copies of a nucleic acid sequence encoding xylose isomerase, wherein the nucleic acid encoding the xylose isomerase is an exogenous nucleic acid. Optionally, the recombinant microorganisms include at least one nucleic acid sequence encoding a xylulose kinase and/or at least one nucleic acid sequence encoding a xylose transporter. The provided recombinant microorganisms are capable of growing on xylose as a carbon source.

摘要: Provided is a method of culturing one or more microorganisms. The method includes culturing one or more microorganisms in a medium comprising crude glycerol at a first concentration level, feeding to the media an additional amount of crude glycerol, once the first concentration of glycerol is reduced to a first threshold level, at a concentration sufficient to achieve the first concentration level, monitoring the crude glycerol concentration until the first concentration level of the crude glycerol is reduced to the first threshold level. The steps may be repeated until a desired microorganism cell density is achieved.

摘要: Provided herein are methods of culturing a microorganism. The method includes providing a container comprising one or more microorganisms and medium, wherein the microorganisms and medium form a start volume. The microorganisms and medium are cultured until the culture reaches a threshold indicator, wherein culturing comprises feeding one or more carbon sources to the culture and wherein the culture is at a threshold volume when the threshold indicator is reached. The method also includes harvesting a portion of the threshold volume to leave a residual volume that is 40% or less of the start volume and adding fresh medium to the container in an amount to return the volume of the culture to the start volume.

摘要: Provided herein are engineered microorganisms comprising one or more genes involved in fatty acid metabolism. For example, provided is an engineered microorganism comprising a first nucleic acid sequence encoding an elongase and a second nucleic acid sequence encoding a desaturase, wherein the first and second nucleic acid sequences are operably linked to promoter. Also provided are methods of making and using the engineered microorganisms. Also provided are microbial oils comprising fatty acids, wherein the fatty acids comprise C20:3(n-6) (di-homo-γ-linoleic acid) and C20:5(n-3) eicosapentaenoic acid (EPA). In addition, provided herein are methods of promoting conversion of saturated fatty acids to unsaturated fatty acids by transforming microorganisms with nucleic acid encoding polypeptides involved in the fatty acid synthesis pathway to generated increased conversion of saturated fatty acids to unsaturated fatty acids.

The methods of recovering oil described herein include contacting a population of microorganisms with one or more enzymes under conditions that cause disruption of the microorganisms, concentrating the disrupted microorganisms, and extracting lipids from the disrupted microorganisms at high temperature in the presence of a salt and in the absence of solvent.

摘要: Provided herein are methods for winterizing oil. The methods include heating the oil to a first temperature and maintaining the oil at the first temperature for a first period of time; reducing the first temperature of the oil after the first period of time to a second temperature over a second period of time, wherein reducing the first temperature produces a first solid fraction and first liquid fraction of the oil; removing the first solid fraction from the oil; reducing the second temperature of the first liquid fraction of the oil over a third period of time to a third temperature, wherein reducing the second temperature of the oil produces a second solid fraction and second liquid fraction of the oil; removing the second solid fraction from the oil; and recovering the second liquid fraction of the oil.
