    摘要: A drug delivery device that includes a capsule for implantation into the body; the capsule further includes a reservoir for containing a substance such as a therapeutic agent, at least one port for allowing the substance to diffuse from or otherwise exit the reservoir, and a nanopore membrane in communication with the capsule at or near the exit port for controlling the rate of diffusion of the substance from the exit port. The device also includes an optional screen for providing structural stability to the nanopore membrane and for keeping the pores of the nanopore membrane clear. One embodiment of the drug delivery device includes an osmotic engine internal to the device for creating fluid flow through the device.

The sealing member is unseated during opening of the poppet to permit fluid to flow into the damping chamber at the inlet side.

    摘要: A method of treating a site of systemic infection which includes administering a therapeutic compound entrapped in liposomes. Also included is a liposomal composition and a method of preparing a liposomal composition for use in concentrating a therapeutic compound to an infected region via the bloodstream. The liposomes, which contain the agent in entrapped form, are composed of vesicle-forming lipids, a vesicle-forming lipid derivatized with hydrophilic biocompatible polymer, and have sizes in a selected size range between 0.07 and 0.2 microns. After parenteral administration, the liposomes are selectively taken up by the infected region within 24-48 hours, for release of entrapped compound into the infected region.

    摘要: A composition and method for treating a wound or surgical incision, by sustained-released delivery of epidermal growth factor (EGF). The composition includes a high-viscosity aqueous dispersion of negatively charged liposomes with liposome-entrapped EGF. The EGF/liposome composition is formed, in one embodiment of the invention, by suspending a lipid mixture typically containing equimolar amounts of neutral and negatively charged phopholipids and cholesterol in a low-conductivity aqueous medium containing EGF and a zwitterionic compound whose isoelectric point is between pH 5.5 and 8.5 to form a gel-like composition. One embodiment of the invention contains EGF entrapped by surface adsorption to the liposomes, for slow release from the composition.
