The microfluidic device can be fluidically coupled to a switching valve to provide for selective control of fluid flow in the chromatography system. In some examples, the microfluidic device may include a charging chamber, a bypass restrictor or other features that can provide for added control of the fluid flow in the system. Methods of using the devices and methods of calculating lengths and diameters to provide a desired flow rate are also described.

The pressure at which the fluid containing the analytes is applied at the inlet of the transfer line is adjusted as the temperature of the column increases in accordance with certain relationships in order to maintain a substantially constant flow rate or velocity for the fluid exiting the column. In this way, the system compensates for changes in the viscosity of the fluid flowing through the column that result from changes in the column temperature.

Valveless conduits permit fluid to be communicated between the sampling device and the first channel, between the first channel and the second channel, and the first channel and the column. In some embodiments, fluid flows in one direction when the analytes are adsorbed and in the opposite direction when analytes are desorbed. In certain embodiments, two different adsorbents are disposed in the second channel to adsorb different types of analytes.

A system and method of measuring the effective temperature inside a sealed container having a headspace is provided. A liquid solvent is added to the container, and a solid compound is added to the liquid solvent to create a saturated solution. Vapor of the saturated solution is allowed to equilibrate in the headspace of the sealed container, and a volume thereof is transferred to a chromatographic column, where chromatographic readings of the equilibrated vapor are taken. A temperature within the sealed container is then calculated based upon the chromatographic readings of the equilibrated vapor, wherein the temperature calculation is based upon the concentrations of the liquid solvent and the solid compound in the equilibrated vapor.

A system and method for extracting headspace vapor is generally disclosed comprising pressurizing a vessel containing headspace vapor with a carrier gas and subsequently venting the sample mixture through an adsorbent trap and out a vent. A flow controller is employed to gradually increase the flow therethrough as the pressure drops as a result of the gradual depletion of headspace vapor in the vessel and, in certain embodiments, the flow controller maintains a constant flow rate. Due to the time saved, in some embodiments, multiple pressurization-venting cycles are implemented to maximize the amount of vapor extracted from the vial. Due to the constant flow rate, in certain embodiments, the pressure decay is monitored and compared to reference values in order to determine whether the sample vessel has a leak or other defect.

Preferably, a heating device allows the tube to be heated.
