The fusion peptides comprise at least one inclusion body tag operably linked to a peptide of interest. Expression of the fusion peptide in a host cell results in a product that is insoluble and contained within inclusion bodies in the cell and/or cell lysate. The inclusion bodies may then be purified and the protein of interest may be isolated after cleavage from the inclusion body tag.

The invention relates to novel spider silk protein analogs derived from the amino acid consensus sequence of repeating units found in the natural spider dragline of Nephila clavipes. More specifically, synthetic spider dragline has been produced from E. coli and Bacillus subtilis recombinant expression systems wherein expressions from E. coli is at levels greater than 1 mg full-length polypeptide per gram of cell mass.

An acid-resistant peptide solubility tag (an “inclusion body tag”) is provided that is effective in producing peptides of interest in an insoluble form. Fusion peptide constructs comprising the inclusion body tag fused to a peptide of interest are provided. An acid cleavable peptide moiety separates the inclusion body tag from the peptide of interest so that acid hydrolysis can be used during subsequent processing steps to separate the tag from the desired peptide of interest. The present inclusion body tag's resistance to acid hydrolysis facilitates easier and cleaner separation of the peptide of interest after acid hydrolysis. Specifically, a ketosteroid isomerase-derived inclusion body tag is provided that has been engineered to be more resistant to acid hydrolysis.

Peptides have been identified that bind with high affinity to dyed hair. Peptide-based hair reagents formed by coupling a dyed-hair-binding peptide to a benefit agent are described. The peptide-based hair reagents include peptide-based hair conditioners and hair colorants. The peptide-based hair conditioners and hair colorants are comprised of a dyed-hair-binding peptide coupled to a hair conditioning agent or a coloring agent, respectively. The peptide-based hair reagents or the dyed-hair-binding peptides may also be used as a protective sealant to improve color retention of dyed hair.
