    摘要: In a method of separating bacteria from a bacteria containing liquid sample by means of a dish-like centrifuge container (21) having an upper opening (25) defined by a radially inwardly extending rim portion (24), the following sequential steps are carried out. The centrifuge container is rotated at a high rotational speed. A volume of a high density component of a density higher than the density of the bacteria is introduced into the centrifuge container, whereupon a volume of a low density component having a density lower than the density of the bacteria is introduced into the container so that a two component gradient separation layer (60) comprising a high density layer (61) and a low density layer (62) is formed within the centrifuge container (21). The liquid sample is introduced through a supply tube (32) into the center of the centrifuge container, and owing to the centrifugal force generated by the rotation of the centrifuge container, the liquid is thrown outwardly from the center of the centrifuge container. The bacteria are forced into the low density component layer (62) and deposited in the interface (63) between the two layers (61) and (62), while the liquid sample is discharged (66) from the upper opening of the centrifuge container through a notch (26). A flushing agent is introduced into the centrifuge container and discharged from the upper opening thereof together with any material adhering at the peripheral surface of the gradient separation layer (60). The centrifuge container (21) is decelerated to a low rotational speed, whereafter a suction pipette (40) is activated and moved from a first position having its tip remote from the inner surface of the peripheral wall of the centrifuge container (21) to a second position having its tip arranged adjacent to said inner surface, while a volume of a transfer liquid is introduced into the centrifuge container so that liquid substance included in the centrifuge container is sucked off therefrom through the pipette tube (41) of the suction pipette (40).

    摘要: Apparatus for separating liquid from solids contained in a liquid-solid slurry includes a centrifugal concentrator which is supplied with feed slurry, and a centrifugal separator to which the output of the concentrator is conducted. The concentrator includes a rotatable central slurry reservoir having outlets discharging slurry under centrifugal force in streams nominally following spiral flow paths. Spirally-curved perforate vanes rotated with the reservoir and intersect respective slurry streams at acute angles. A fractional portion of the fluid in the slurry is removed by filtration as each stream intersects and then flows along a spirally-curved perforate vane, thereby to provide concentrated slurry, which is supplied to the separator. The separator includes a high-speed rotatable carrier, which carries eccentrically-disposed filter baskets independently rotatable at low speeds about their own axes. Each basket includes a base plate, and spirally-curved perforate vanes mounted on and extending outwardly from a central region of the base plate. A reservoir having restrictive outlets for discharge adjacent to the inner ends of respective vanes is mounted in juxtaposition to the central region of each basket, for rotation with the basket. Concentrated slurry from the concentrator is supplied to each basket reservoir, is discharged from the reservoir outlets, and flows radially outwardly of the carrier axis of rotation under the application of centrifugal force, when the carrier is rotated. With the concurrent flow rotation of each basket about its axis, the slurry is filtered substantially over the full length of each basket vane, with liquid passing through the vane and solids being retained on the vane surface, for discharge therefrom separately from the filtrate.

    摘要: An improved centrifuge assembly comprising a channeled rotor assembly and a fluid container disposed in the channel, whereby the centrifugal separation effects in the fluid container are determined by the geometry of the channel in the rotor. This arrangement is particularly useful for two-stage blood platelet separation. The fluid container is preferably formed from semirigid plastic material and is considered a disposable item to be discarded after a single use. The rotary assembly preferably includes a removable filler piece or center piece formed from a single piece of material, such as rigid plastic, as by machining or molding, and having therein an open-topped channel having dimensions appropriate to receive the semirigid container, which is suitably curved and placed in the channel. Fluid connections are provided from each end of the container and an intermediate point to an axially located multichannel rotating seal. The connections lie in a plurality of radial slots in the filler piece. The channel is divided into two distinct portions, the first portion of the channel being circular with a constant radius, and the second portion being spiral and having a plurality of radiuses, each measured from a different center, the spiral increasing radially outward from its juncture with the first stage. The intermediate fluid connection is established near the junction of the first stage and the second stage. The spiral portion of the channel and container may have an outward slope from bottom to top.
