The L-fucose α1→6 specific lectin of the present invention is characterized in that: (1) the lectin is extracted from a basidiomycete or an ascomycete; (2) the lectin has a molecular weight by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) of 4,000 to 40,000; and (3) the lectin has an affinity to an L-fucose α1→6 sugar chain, the affinity being represented by an association constant of 1.0×104M−1 or more (at 25 degrees C.). The lectin can be used for detecting an L-fucose α1→6 sugar chain specifically, and is effective for the purification of an L-fucose α1→6 sugar chain or a non L-fucose α1→6 sugar chain.

摘要: As a result of investigating the optimum conditions of methods for immobilizing proteins that interact with sugar chains onto a substrate, it was revealed that coating the surface of a slide glass with GTMS enables immobilization at a higher S/N ratio than conventionally possible. Moreover, by using a substrate to which a rubber with a number of holes was affixed to form a number of reaction vessels, and further by spotting lectins onto the substrate and washing with PBST, the weak interactions between sugar chains and lectins were successfully detected with improved sensitivity. In addition, by introducing an evanescent excitation-type scanner, it became possible to detect the interactions between lectins and sugar chains without washing away the probe solution.

摘要: In the case where many interactional substances are immobilized on the front surface side of a light transmitting plate capable of generating evanescent waves on the front surface, and where the fluorescence levels in the interactions between the interactional substances and a fluorescence labeled test substance are detected in a liquid layer according to a conventional method, the detection sensitivity and S/N ratio are low due to the background noise. Furthermore, it is necessary to perform a gel filtration purification step for the purpose of removing an excessive amount of the fluorescence dye used for labeling a test substance before the interactions between the fluorescence labeled test substance and the interactional substances are performed. To solve these problems, this invention proposes the above-mentioned method for detecting the evanescent wave excited fluorescence comprising the step of adding a black colorant, for example, a pigment based colorant mainly containing fine particles such as Bokuju.

摘要: A thermal head including a protection layer having mutually opposed first and second surfaces, said first surface having a flat or protruded printing surface which is brought into contact with a heat sensitive record medium, a heat generating section including resistors and electrodes connected to the electrodes and provided on said second surface of the protection layer, and a reinforcing member made of a low melting pint glass and provided on a side of the heat generating section remote from the protection layer. The reinforcing member improves a mechanical strength of the thermal head. The reinforcing member made of a glass also serves as a heat storage member, and thus a thermal property of the thermal head is improved. The reinforcing member may be formed by an aggregate of ceramic particles. The reinforcing member may contain a heat storage layer made of a low melting point glass and a heat conduction layer provided on the heat storage layer.
