A polyacrylamide gel comprising acrylamide and diacrylylpiperazine. A method for providing a polyacrylamide gel comprising employing a catalyst system which comprises dimethylpiperazine, sodium thiosulfate or a mixture thereof and ammonium or potassium persulfate. The use of the gel as the matrix in a silver staining procedure for the detection of biomaterials, provides for the obtainment of reduced background staining.

A strip gel and a slab gel are combined in a single gel enclosure or on a single gel backing for two-dimensional electrophoresis. The enclosure is comprised of a single pair of support plates, and a removable electrically insulating region is included to separate the gels. The region is occupied either by air or an electrically nonconductive material which can be removed without disturbing either gel. In use, a sample is loaded onto one end of the strip gel and an electric potential is imposed across the strip gel for the first dimension separation while the strip gel is electrically insulated from the slab gel. The insulating region is then removed and the two gels placed in electrical contact for the second dimension separation.

Amine-acryloyl and -methacryloyl derivatives and 1,6-heptadiene-4-ol can be used to provide crosslinked polyacrylamide gels which are useful in protein and nucleic acid separation and detection. The gels crosslinked with the compounds of the present invention exhibit much less background silver staining than previously known acrylamide gels.

Polyacrylamide gels containing chaotropic agents such as urea as solubilizing agents for protein or nucleic acid separations are rendered stable for prolonged periods of time by the use of certain diacrylyl compounds with tertiary amide groups as cross linking agents. These compounds have the general structure ##STR1## where the R groups are generally saturated hydrocarbon groups and R.sup.4 through R.sup.9 may also be hydrogen. When used as cross linkers, these compounds have the further advantage of preventing urea crystallization at lowered temperatures.
