The invention provides an improved instrument for automation of solid phase extraction chemistries typically used in biotechnology labs. The instrument includes a mechanism for transferring samples dissolved in a liquid from initial containers to reaction columns that are used to perform solid phase extractions. Samples, reaction columns and collection containers are in microtiter plate format or tubes that are on 18 millimeter centers. The transfer system is automatically cleaned after use in preparation for the next use. A dispense manifold is used to dispense various reagents into the reaction columns. Pressure differential is used to move reagents through reaction columns. A sliding cover is used to divert the reagent exiting reaction columns to waste or allowing collection of sample as it exits outlets of reaction columns. Samples are automatically collected in microtiter plates or in individual tubes or vials.

Flow control mechanisms control the direction and flow rate of synthesis reagent through one or more synthesis reaction vessels for automated solid phase synthesis. Selectable, known, and reproducible positive or negative pressure differentials (−5 to +10 psi) accomplish controlled, bidirectional (forward and reverse) flow of synthesis reagents through synthesis media contained within the reaction vessels. Venturi-based vacuum apparatus, valves, electronic pressure regulators and compound digital pressure gauge, can be added to automated solid phase synthesis instruments to provide, control, and monitor known, selectable, reproducible negative and positive pressures to one or both valve sealable and un-sealable ends (inlets and outlets) of the reaction vessel as needed to generate and reverse said pressure differentials between the opposite ends of said synthesis reaction vessels, yielding controlled forward and backward flows of synthesis reagents through the synthesis media.

A mechanism for securing tubes in a fixed position is described wherein a body to which a tube is to be fixed has at least one smooth bore hole extending therethrough. A tube has an inner diameter accommodating fluid flow and an outer diameter passing through the smooth bore hole in slip fit relation with the smooth bore of the hole. A threaded hole with helical grooves is parallel to the smooth bore hole and located such that its grooves intersect the diameter of the smooth bore hole. A set screw made of a tougher material than the tube has threads that will seat in the threaded hole in a manner such that advancing the set screw scratches the outer diameter of the tube to a depth wherein the set screw retains the tube in place without deformation of the inner diameter of the tube whereby fluid flow in the tube is not affected by advancement of the set screw while the tube is retained in place by the set screw. The invention can connect tubes in all sorts of patterns with many center-to-center tube distances.

The system and methods include a mechanism for controlling the pressure differential across the reaction vessels that contain the solid support used in solid phase synthesis. Reaction vessels are held in a holder that provides a sealable chamber at the outlet ends of the reaction vessels. The rotor containing the reaction vessels is placed within a sealable chamber. The sealable chamber is fitted with a means for engaging the rotor and draining the reaction vessels to waste. The sealable chamber is also fitted with a means for engaging the rotor to drain the reaction vessels at a slower, variable rate.
