摘要: Compositions, systems and related methods for delivering a supercharged protein or a complex of a supercharged protein and therapeutic agent (e g, nucleic acid, peptide, small molecule) to cells are disclosed. Superpositively charged proteins may be associated with nucleic acids (which typically have a net negative charge) via electrostatic interactions. The systems and methods may involve altering the primary sequence of a protein in order to “supercharge” the protein (e g, to generate a superpositively-charged protein). The compositions may be used to treat proliferative diseases, infectious diseases, cardiovascular diseases, inborn errors in metabolism, genetic diseases, etc.

Such systems and methods include the use of supercharged proteins. For example, superpositively charged proteins may be associated with nucleic acids (which typically have a net negative charge) via electrostatic interactions. In some embodiments, such systems and methods involve altering the primary sequence of a protein in order to “supercharge” the protein (e.g., to generate a superpositively-charged protein). In some embodiments, complexes comprising supercharged proteins and one or more agents to be delivered are useful as therapeutic agents. In some embodiments, complexes and/or pharmaceutical compositions thereof are administered to a subject in need thereof. The inventive complexes or pharmaceutical compositions thereof may be used to treat proliferative diseases, infectious diseases, cardiovascular diseases, inborn errors in metabolism, genetic diseases, etc.

摘要: A drop height recorder apparatus includes a housing suitable for packaging for shipment from an origin point to a destination point, a triaxial accelerometer provides three signals to corresponding high gain and low gain channels. The three high gain channel signals are full wave rectified to change any negative signals to positive signals for summing into a single high gain "zero G" (free-fall) channel. AN analog to digital converter is connected to the three low gain channels, the single high gain channel, and to auxiliary sensors for digitizing the analog signals. A recorder has a programmable controller connected to the ADC for setting the digital sampling rate. The digitized low gain channel signals are passed through a digital trigger threshold comparator for comparison with a reference signal for generating event detecting trigger signals on the way to a delay memory which also receives the digitized high gain zero G signal and auxiliary signals. In response to a trigger signal the programmable controller reads out of the delay memory preselected numbers of pre-trigger and post-trigger data as an event sample either for a data processing memory for processing the signals for desired information and storage in a storage memory or for a storage memory. A pre-selected number of event waveform signals together with time, date, and auxiliary data signals are stored in the storage memory. After recovery of the instrument, a personal computer can be attached for reading out the stored event waveforms from which the time of free-fall occurring during each event is determined and the drop height in inches computed. The personal computer is used prior to shipment to set the program parameters and operation modes in the programmable controller.

摘要: A monitor-actuator pilot valve operable to close a safety valve in a fluid flow system reponsive to low line pressure or electrical failure. The safety valve is held open by pressure supplied into an actuator cylinder from the line protected by the safety valve. The pilot valve includes a first flow passage communicating with line pressure, a second flow passage communicating with the safety valve actuator cylinder, a third exhaust flow passage to the atmosphere, and a fourth flow passage to a fluid pressure reset. A valve assembly in the pilot valve responds to a low pressure in the first passage or electrical failure to connect the second and third passages for exhausting control pressure from the safety valve actuator cylinder to close the safety valve. An electromagnet on the pilot valve releases the valve assembly in response to a power failure allowing the safety valve to close. A reset assembly in the valve body manually or hydraulically operates the valve assembly reopening the pilot valve. The low pressure response mechanism is reset hydraulically or manually. The electrical response mechanism is reset only manually.
