    摘要: Provided is a method of producing one or more sesquiterpene compounds comprising: contacting an acyclic FPP precursor with a polypeptide having terpene synthase activity, wherein the polypeptide comprises an amino acid sequence that has at least 55% sequence identity to SEQ ID NO: 1, to produce one or more terpenes selected from the group consisting of isovalencene, spirovetiva-1(10),7(11)-diene and valencene or derivatives thereof, or mixture of sesquiterpenes comprising one or more of isovalencene, spirovetiva-1(10),7(11)-diene and/or valencene; and optionally isolating the one or more terpenes or the mixture. Also described is a nucleic acid derived from Vetiveria zizanoides encoding a polypeptide having sesquiterpene synthase activity, a polypeptide that can be used to produce one or more sesquiterpenes or a mixture of sesquiterpenes comprising one or more of isovalencene, spirovetiva-1(10),7(11)-diene and/or valencene, and a non-human organism or cell comprising the nucleic acid or comprising an expression vector comprising the nucleic acid.

The method allows the fully biochemical synthesis of terpene compounds, like for example copalol and labdendiol, and derivatives thereof, which serve as valuable intermediates for the production of perfumery ingredients, such as, for example, ambrox. Also provided are novel fully biochemical multistep processes for the production of such compounds as well as novel phosphatase enzymes and mutants and variants derived therefrom.

    摘要: Described herein are methods of producing (+)-manool, the methods including: contacting geranylgeranyl diphosphate with a copalyl diphosphate (CPP) synthase to form a (9S, 10S)-copalyl diphosphate and contacting the CPP with a sclareol synthase enzyme to form (+)-manool and derivatives thereof. Also described herein are nucleic acids encoding CPP synthases and sclareol synthases for use in the methods. Further described herein are expression vectors and non-human host organisms and cells including nucleic acids encoding a CPP synthase and a sclareol synthase as described herein.
