    Abstract: Gasket assemblies and related system and methods. An apparatus includes a system, a flow cell, and a plurality of gasket assemblies. The system includes a flow cell interface and the flow cell has one or more channels. Each channel has a first channel opening and a second channel opening. The first channel openings are positioned at a first end of the flow cell and the second channel openings are positioned at a second end of the flow cell. A gasket assembly coupled at each second channel opening. Each gasket assembly includes an adhesive stack and a gasket. The adhesive stack includes a first side bonded to the gasket and a second side bonded to the flow cell. The flow cell interface is engagable with the corresponding gaskets to establish a fluidic coupling between system and the flow cell.

    Abstract: Library preparation systems and methods are disclosed. In an implementation, a modular bay for preparing a library of samples for sequencing, the modular bay comprising a contact dispenser and a working area comprising a working plate receptacle adapted to receive a working plate, a thermocycler, a magnet, and a drawer. The drawer comprising a consumables area adapted to receive a sample plate adapted to contain a sample, and a plurality of consumables for interacting with the sample in the working plate. The contact dispenser is linearly movable in a longitudinal direction between the consumables area and the working area, such that the contact dispenser is configured to (i) move the sample from the sample plate in the consumables area to the working plate in the working area, and (ii) move the plurality of consumables between the consumables area and the working plate in the working area.

    Abstract: A method includes engaging a well of a cartridge with a flow sensor of an instrument. The cartridge includes: a rotary valve including a rotatable port and a center port; the well in fluid communication with a channel, the channel including a channel port that the rotatable port is to align to in order to receive fluid from the well; and a flowcell including an inlet gasket in fluid communication with the center port. A source of pressurized air is connected to the flow sensor in order to establish a mass flow of air through a flow path. The flow path extends through one of the flow sensor, the channel, the rotary valve, and the flowcell. The mass flow of air through the flow path is measured with the flow sensor. It is determined if there is one of an air leak and an air blockage within the flow path.

In accordance with an implementation, an apparatus includes a first reagent cartridge, a second reagent cartridge, and a system. The system includes a sipper manifold assembly having a sipper, a queue to carry the first reagent cartridge and the second reagent cartridge, a cartridge moving assembly, and a cartridge receptacle assembly. The cartridge moving assembly includes a gantry, a carriage actuator, and a carriage. The carriage is coupled to the gantry and the carriage actuator is to move the carriage relative to the gantry. The cartridge receptacle assembly has a cartridge receptacle to receive the first reagent cartridge or the second reagent cartridge. The carriage is to move the first reagent cartridge from the queue and position the first reagent cartridge into the cartridge receptacle.
