The coloring sensitivity of the dry-type analytical element does not increase substantially even under a fluorescent light, and thereby, an accurate measured value can easily be obtained.

A multilayer analytical element for measuring enzyme activity in a liquid sample such as a body fluid which is less affected by intrinsic pyruvic acid contained in the liquid sample is disclosed. The analytical element comprises a support, a reagent layer, an optionally provided other layer, and a porous spreading layer. The reagent layer and/or the optionally provided other layer contain pyruvic acid oxidase. The characteristic feature of the analytical element lies in that the spreading layer has a specific void volume in the range of 3-15 .mu.l/cm.sup.2.

An analysis film for assaying transaminase activity comprises a reagent layer containing pyruvic acid oxidase, a combination of a hydrogen donor and a coupler--which is capable of forming a cationic dye--and an anionic polymer. The cationic dye formed by the reaction with a color indicator composition for detecting hydrogen peroxide is fixed with the anionic polymer, thereby mobilization and diffusion of the cationic dye being prevented so that the analysis film achieves high detection sensitivity and high accuracy.

A quantitative analysis film comprising a reagent layer comprising a cationic dye-forming color indicator for detecting hydrogen peroxide and an anionic polymer. By the use of such a color indicator and an anionic polymer in combination, a cationic dye is formed; the formed cationic dye is then bound and fixed to the anionic polymer to provide an intensified color. A method for the colorimetric assay of hydrogen peroxide or a precursor thereof with high sensitivity and high efficiency is also disclosed.

The color-former is a precursor for forming a dye in the presence of hydrogen peroxide and peroxidase. The reagent composition of the invention is improved in storage stability and has a wider determination range. Also provided is a dry analysis element composed of reagent layer containing the inventive reagent composition.

A method for correcting an instrumental error due to wavelength error of a spectroscope of an optical analyzer for measuring an optical density reflected from or transmitted through a dry analysis element. One standard color plate having a standard optical density OD.sub.ST is measured by an optical analyzer to be corrected to obtain an measured value OD.sub.M. A measured optical density OD.sub.S of the element applied with a sample through the optical analyzer to be corrected is corrected by using a ratio of OD.sub.M /OD.sub.ST to obtain a corrected measured value OD.sub.C of the sample. The standard color plate to be used contains an indicator dye contained in the non-reacted dry analysis element or a dye having an absorption spectrum same as or similar to that of the indicator dye in the measurement wavelength range. Another correction method in consideration of a layer coefficient of a dry analysis element is also provided.
