    摘要： This invention concerns a family of chimeric antibodies with high affinities to a high molecular weight, tumor-associated sialylated glycoprotein antigen (TAG-72) of human origin. These antibodies have (1) high affinity animal VH and VL sequences which mediate TAG-72 binding and (2) human CH and CL regions. They are thought to produce significantly fewer side-effects when administered to human patients by virtue of their human CH and CL antibody domains. The nucleotide and amino acid sequences of VH&agr;TAG VH, CC46 VH, CC49H, CC83 VH, and CC92 VH, and CC49L, CC83 VL, and CC92 VL idiotype sequences are disclosed, as well as in vivo methods of treatment and diagnostic assay using these chimeric antibodies.

    摘要： Rapid screening of a large number of organisms for variants producing particular products is accomplished using membrane plates having a predetermined molecular weight cutoff and which divide a container into two chambers. High density cellular lawns are employed in conjunction with labeled antibodies for the product of interest mixed in soft agar. The cells are subjected to viral lysis and after the plaques have reached maturity, the plates are immersed in buffer. Residual soluble labeled antibody diffuses from the agar, and immunoprecipitate of the product and the antibody may then be detected.

    摘要： An improved horizontal slab gel electrophoresis apparatus having compartments for vertical wicks at each end of a removable horizontal trough. Adjacent to the vertical wicks are vertical buffer compartments with built in electrodes. A partition separates the vertical wicks from the buffer compartment and includes a slot with a silicone plug which can be removed after curing or hardening of the vertical wicks to permit communication between the buffer and the gel slab. Flow adapters and an interconnecting tube permit constant circulation of the buffer to maintain equal volume and pH. The horizontal trough has slots for insertion of combs to form up to seventy-five sample wells and additional slots at each end for sealing the end of the horizontal slab when the gel slab is removed. The apparatus includes coils in a horizontal bed or plate beneath the trough for cooling the gel slab during a run and levelling screws for levelling the horizontal trough before pouring the gel slab.
