    Abstract: Methodologies for labeling the epigenetic modification 5-hydroxymethyl-cytosine (5hmC) along a DNA molecule, and for determining a presence or a level of this epigenetic modification based on a ratio of fluorescence intensity of a labeled DNA sample to absorption intensity of the DNA sample at 260 nm are disclosed. Related compositions and reagents, and methods of preparing same are also disclosed.

    Abstract: Methods of detecting methylated CpG are provided. Accordingly, there is provided a method of determining CpG methylation status in a DNA sample, the method comprising: (a) subjecting the DNA sample to bisulfite conversion; (b) amplifying said DNA sample following said (a) to obtain an amplified DNA sample; (c) labeling CpG sites in said amplified DNA sample with a label to obtain a labeled DNA sample; (d) contacting said labeled DNA sample on an array comprising a plurality of probes for said DNA under conditions which allow specific hybridization between said plurality of probes and said DNA; and (e)detecting said hybridization, wherein an amount of said label is indicative of the CpG methylation status in said DNA sample.

    Abstract: Methods and arrays for identifying the cell or tissue origin of DNA are provided. Accordingly there is provided a method of identifying DNA having a methylation pattern distinctive of a cell or tissue type or state comprising: labeling an epigenetic modification of interest in a DNA sample with a label; contacting said sample on an array comprising a plurality of probes for said DNA under conditions which allow specific hybridization between said plurality of probes and said DNA; and detecting said hybridization, wherein an amount of said label is indicative of the cell or tissue type or state, wherein the method is effected in the absence of amplification of said DNA.

    Abstract: A method of detecting an epigenetic DNA modification is described herein, the method comprising: attaching to DNA a labeling agent selective for the DNA modification; contacting an aqueous solution comprising the DNA with at least one addressable region on a surface of a substrate, wherein the addressable region comprises a positively charged substance capable of attaching the DNA to the surface, and the addressable region is surrounded by a hydrophobic region; and determining an amount of labeling agent in the addressable region. Further described herein is a kit comprising a substrate comprising discrete hydrophilic regions separated by a hydrophobic region, and a labeling agent selective for an epigenetic DNA modification; as well as an article of manufacture comprising a substrate comprising a discrete regions separated by a hydrophobic region, a positively charged substance and DNA attached thereto, the DNA comprising such a labeling agent.
