The present invention relates to methods and compositions for the preservation of organs. The inventors showed that therapeutic intervention during cold ischemia could significantly alter outcome, suggesting active mechanisms taking place. More particularly, the inventors demonstrated that Unfolded Protein Response could be a critical pathway underlying the relationship between cold ischemia time and graft outcome, highlighting the potential for UPR-based therapeutics to improve transplantation efficiency. In particular, the present invention relates to an activator of PERK-ATF4 pathway, an inhibitor of the ATF6 pathway and an inhibitor of the RNase activity of IRE1α for use in the maintain of organ viability during cold ischemia.

A method for cryopreservation of adherent cells attached to a substrate and a method for identifying one or more extracellular matrix (ECM) components and/or matricellular proteins that improves viability and retention of the cryopreserved cells on the substrate. The method for cryopreservation includes treating a substrate with at least one ECM component and/or adding at least one matricellular protein to the substrate or cell culture media, plating the cells on the treated substrate, and cryopreserving the cells. One or more ECM components and/or matricellular proteins that improves cell viability and retention can be identified by evaluating the cells that have been thawed from the cryopreservation temperature to determine cell viability and retention.

Surgical techniques typically employed for vein harvest—stretching the vein, placing the vein in low pH, solutions, and the use of toxic surgical skin markers—are shown here to cause injury. The invention therefore provides for non-toxic surgical markers than also protect against stretch-induced loss of functional viability, along with other additives. The present invention relates to a novel method for the preparation of a pooled or mixed population of cryopreserved cells (e.g. hepatocytes). In particular, the invention entails the rapid thaw of cells (e.g. hepatocytes), donated from a single individual, which are mixed to create a heterogeneous population and then cryopreserved. The invention also concerns preparations of multi-cryopreserved cells to increase viability prior to immediate use. The process entails reducing exposure to chemical and physical stresses to increase the resultant number of viable cells.
