Disclosed is an Aspergillus fumigatus AFAP1 polypeptide and DNA (RNA) encoding such AFAP1 polypeptide. Also provided is a procedure for producing such polypeptide by recombinant DNA techniques and a procedure for generating antibodies against the polypeptide. Also disclosed is a method of using such polypeptide and the antibodies against it for the diagnosis of deep infections of Aspergillus fumigatus by detecting the presence of the specific antibodies as well as the AFAP1 protein antigen in clinical specimens taken from suspected patients. Also provided are methods of using the AFAP1 DNA(RNA) or protein sequence to identify and to clone its homologous genes from other Aspergillus species. Therefore, the identification of AFAP1 homologous genes from other pathogenic fungi are made possible with this invention. Also described is a therapeutic regimen using the antibodies against Aspergillus infection. Also provided is a method of immunization against the infection of Aspergillosis.

The present invention provides a koji mold having increased protease activity and peptidase activity relative to a parent strain, a method of breeding the koji mold, and a method of manufacturing a flavor enhancer using the koji mold. More specifically, the present invention provides: (1) a koji mold having increased protease activity and peptidase activity relative to a parent strain obtained by transformation using a protease gene and a peptidase gene. (2) a method of breeding the above koji mold which comprises transforming a parent strain of koji mold using a protease gene and a peptidase gene, and then selecting a transformant having higher protease activity and peptidase activity relative to a parent strain. (3) a method of manufacturing a flavor enhancer which comprises allowing a culture product of the above koji mold to act on a protein.

Described herein are methods for increasing the amount of protein secreted by a cell. In one case, a cell is provided which contains a heterologous nucleic acid encoding a protein having unfolded protein response modulating activity and a heterologous nucleic acid encoding a protein of interest to be secreted. In one case, the protein having unfolded protein response modulating activity is selected from the proteins selected from the group consisting of HAC1, PTC2 and lRE1. The protein of interest can be any secreted protein such as a therapeutic or an industrial enzyme. For example the protein can be selected from the group consisting of lipase, cellulase, endo-glucosidase H, protease, carbohydrase, reductase, oxidase, isomerase, transferase, kinase, phosphatase, alpha-amylase, glucoamylase, lignocellulose hemicellulase, pectinase and ligninase.
