The present invention provides methods, media and compositions capable of modulating the differentiation of stem cells. Applicants have discovered that agonists of lysophospholipid receptors and ligands of class III tyrosine kinase receptors are useful in preventing the spontaneous differentiation of stem cells. The ligands and agonists may be used alone, or in combination where they have a synergistic effect. Also provided are cells produced using the methods and media, and methods of treating stem cell related diseases using the compositions described herein. Methods of identifying compounds useful in finding other agents useful in the modulation of stem cell differentiation are also disclosed.

The invention relates generally to methods of influencing central nervous system cells to produce progeny useful in the treatment of CNS disorders. More specifically, the invention includes methods of exposing a patient suffering from such a disorder to a reagent that modulates the proliferation, migration, differentiation and survival of central nervous system cells. These methods are useful for reducing at least one symptom of the disorder. The reagent used is platelet-derived growth factor (PDGF), vascular endothelial growth factor (VEGF), a combination of both, a PDGF agonist or a VEGF agonist.

Polypeptides comprising essentially the extracellular domain of a human PDGF receptor fused to dimerizing proteins, the portion being capable of binding human PDGF or an isoform thereof, are also disclosed. The polypeptides may be used within methods for determining the presence of and for purifying human PDGF or isoforms thereof. Pharmaceutical and diagnostic compositions utilizing the polypeptides are also disclosed.

Discoveries are disclosed that show particular aspects of recombinant DNA technology can be used successfully to produce a hitherto unknown type of human Platelet-Derived Growth Factor (PDGF) receptor protein (see genetic map, figure 2) free of other PDGF receptors. These proteins can be produced from DNA segments in cells in various functional forms. These forms variously enable biochemical and functional studies of these novel receptors as well as production of antibodies. Means are described for determining the level of expression of genes for specific types of PDGF receptor proteins, for example, by measuring mRNA in cell with PDGF receptor type-specific DNA probes or by measuring antigene in biological samples with type-specific antibodies.

The present invention provides methods and systems to predict and diagnose inflammatory bowel disease (IBD) and subtypes such as ulcerative colitis (UC) and Crohn's disease (CD) by detecting the presence, absence, level, and/or genotype of one or more sero-genetic-inflammation markers. Advantageously, with the present invention, it is possible to provide a diagnosis of IBD versus non-IBD, to rule out IBD that is inconclusive for CD and UC, and to differentiate between CD and UC with increased accuracy.
