    摘要: An object of the present invention is to prepare individual microorganisms or a microflora composition derived from land or seafloor soil in which contamination of land or seafloor soil-derived substances is reduced. The present invention provides [1] a method for preparing, from a land or a seafloor soil samples, microorganisms containing a reduced level of land or seafloor soil-derived substances that inhibit nucleic acid analysis, which comprises (1) the step of disposing a suspension of a land or seafloor soil sample containing microorganisms and land or seafloor soil-derived substance that inhibit nucleic acid analysis as an electrolyte on a surface of a substrate at least a part of which constitutes an electrode, and applying a constant potential to the electrode to attach at least a part of the microorganisms to the surface of the substrate, and (2) the step of applying a high-frequency wave potential to the electrode to detach the microorganisms attached to the surface of the substrate in a viable state, and wherein the constant potential applied in the step (1) is greater than -0.5 V but not greater than-0.2 V (vs. Ag/AgCl), or greater than +0.2 V but not greater than +0.4 V (vs. Ag/AgCl), and the high-frequency wave potential applied in the step (2) has a frequency in the range of from 1 kHz to 20 MHz, and a potential range of ±0.1 V (vs. Ag/AgCl) or smaller.

    摘要: The present invention includes devices, systems, and methods for assaying cells using cell-substrate impedance monitoring. In one aspect, the invention provides cell-substrate impedance monitoring devices that comprise electrode arrays on a nonconducting substrate, in which each of the arrays has an approximately uniform electrode resistance across the entire array. In another aspect, the invention provides cell-substrate monitoring systems comprising one or more cell-substrate monitoring devices comprising multiple wells each having an electrode array, an impedance analyzer, a device station that connects arrays of individual wells to the impedance analyzer, and software for controlling the device station and impedance analyzer. In another aspect, the invention provides cellular assays that use impedance monitoring to detect changes in cell behavior or state. In some preferred aspects, the assays are designed to investigate the affects of compounds on cells, such as cytotoxicity assays. In other preferred aspects, the assays are designed to investigate the compounds that effect IgE-mediated responses of cells to antigens.

    摘要: A method for preparing animal cells with good adhesion and proliferation properties which are capable of proliferation is provided. the method permits the detachment of cultured cells without damaging the cells. A method for preparing a sheet of animal cells such as skin cells which are capable of proliferation is also provided. A method for preparing animal cells capable of proliferation, comprising the steps of (1) culturing animal cells on a substrate at least one portion of which is an electrode, and (2) applying a high-frequency wave potential to the electrode to detach the cells that have adhered to the substrate surface through culturing. The high-frequency wave potential is of a frequency falling within a range of 1 KHz to 10 MHz with a potential falling within a range of ± 1.0 V (vs. Ag/AgCl) or less. The culture medium during separation does not contain calcium or magnesium.
