Described are recombinant microorganisms characterized by having phosphoketolase activity, having a diminished or inactivated Embden-Meyerhof-Parnas pathway (EMPP) by inactivation of the gene(s) encoding phosphofructokinase or by reducing phosphofructokinase activity as compared to a non-modified microorganism and having a diminished or inactivated oxidative branch of the pentose phosphate pathway (PPP) by inactivation of the gene(s) encoding glucose-6-phosphate dehydrogenase or by reducing glucose-6-phosphate dehydrogenase activity as compared to a non-modified microorganism. These microorganisms can be used for the production of useful metabolites such as acetone, isobutene or propene.

A process for inhibiting a mammalian cancerous cell or virally infected cell includes providing a Trichomonas vaginalis purine nucleoside phosphorylase enzyme or a tail mutant purine nucleoside phosphorylase enzyme in proximity to the mammalian cancerous cell or the virally infected cell and exposing the enzyme to a purine nucleoside phosphorylase enzyme cleavable substrate to yield a cytotoxic purine analog. The process includes introducing to the cell a vector containing the phosphorylase enzyme, or a DNA sequence coding for the same and delivering to the cell an effective amount of the substrate such as 9-(²-D-arabinofuranosyl)-2-fluoroadenine (F-araA).

Anti-Dandruff Agents A novel anti-dandruff composition comprising spirofuranone-lactam tetramic acid or derivatives thereof, and optionally at least one biologically derivable meroterpene. The spirofuranone-lactam tetramic acid is preferably a bio-active heterospirocyclic secondary metabolite of Aspergillus, such as pseurotin A. The biologically derivable meroterpene may be selected from fumagillin, fumagillin derivative, chlovalicin, or ovalicin. Use of said agents as anti-dandruff actives in anti-dandruff compositions, particularly shampoos and conditioners is also provided. The active is particularly effective against Malassezia yeasts and Malassezia furfur which may cause dandruff. A method of obtaining the anti-dandruff actives from culturing of Peyronellaea sp. strain RKDO347 is also described.

The present invention provides biosynthetic methods for production of caffeine comprising: providing guanine, a guanine deaminase, at least one methyl transferase, and a methyl donor; contacting the guanine with the gtheuanine deaminase to produce xanthine; contacting the xanthine with the methyl transferase and a methyl donor, under conditions wherein the xanthine is methylated, to produce a monomethylxanthine; contacting the monomethylxanthine with the methyl transferase and a methyl donor, under conditions wherein the monomethylxanthine is methylated, to produce a dimethylxanthine; and contacting the dimethylxanthine with the methyl transferase and a methyl donor, under conditions wherein the dimethylxanthine is methylated, to produce caffeine (i.e., 1,3,7-trimethylxanthine).

The invention provides compositions and methods for the recombinant production of cytokinin synthase, host cells and transformants that include the cytokinin synthases, as well as compositions and formulations that include the disclosed cytokinin synthase.
