The present application relates to compositions and methods for treating skin disorders, ,dry skin, and protection of skin in inflammatory events. The present application more particularly discloses the identification of new genes and metabolic pathways involved in skin disorders, which provide novel targets and approaches for treating said disorders and for screening biologically active compounds. The present invention also provides various products and constructs, such as probes, primers, vectors, recombinant cells, which can be used to implement the above methods. The invention may be used to detect or treat various skin disorders, particularly dry and inflammatory skin disorders, in various subjects, including mammalian subjects, particularly human beings.

The present invention relates to a mutant, non-naturally occurring or transgenic tobacco plant cell comprising: (i) a polynucleotide comprising, consisting or consisting essentially of a sequence encoding a functional nicotine N-demethylase and having at least 95% sequence identity to SEQ ID NO:2; (ii) a polypeptide encoded by the polynucleotide set forth in (i); (iii) a polypeptide comprising, consisting or consisting essentially of a sequence encoding a nicotine N-demethylase and having at least 95% sequence identity to SEQ ID NO:3; or (iv) a construct, vector or expression vector comprising the isolated polynucleotide set forth in (i), and wherein the expression or activity of said nicotine demethylase is reduced as compared to a control tobacco plant cell in which the expression or activity of said nicotine demethylase has not been reduced.

The present invention relates to a method for the production of a diterpene or a glycosylated diterpene, which method comprises: a. fermenting a recombinant microorganism in a suitable fermentation medium, wherein the microorganism comprises one or more nucleotide sequence(s) encoding: a polypeptide having ent-copalyl pyrophosphate synthase activity; a polypeptide having ent-Kaurene synthase activity; a polypeptide having ent-Kaurene oxidase activity; and a polypeptide having kaurenoic acid 13-hydroxylase activity and whereby expression of the nucleotide sequence(s) confer(s) on the microorganism the ability to produce at least steviol, whereby a diterpene or glycosylated diterpene is produced extracellularly in the fermentation medium; and b. recovering the diterpene or glycosylated diterpene from the fermentation medium.
