摘要: A method for replicating and amplifying a target nucleic acid sequence is described. A method of the invention involves the formation of a recombination intermediate without the prior denaturing of a nucleic acid duplex through the use of a recombination factor. The recombination intermediate is treated with a high fidelity polymerase to permit the replication and amplification of the target nucleic acid sequence. In preferred embodiments, the polymerase comprises a polymerase holoenzyme. In further preferred embodiments, the recombination factor is bacteriophage T4 UvsX protein or homologs from other species, and the polymerase holoenzyme comprises a polymerase enzyme, a clamp protein and a clamp loader protein, derived from viral, bacteriophage, prokaryotic, archaebacterial, or eukaryotic systems.

摘要: Neonatal seizure is different from adult seizure, and many antiepileptic drugs that are effective in adults often fail to treat neonatal seizure. Gluconic acid, a natural organic acid enriched in fruits and honey, and the glucose oxidase enzyme, is shown herein to potently inhibit neonatal epilepsy both in vitro and in vivo. Sodium gluconate is shown to inhibit epileptiform burst activity in cell cultures and protect neurons from kainic acid-induced cell death. Sodium gluconate also inhibited epileptiform burst activity in brain slices in a manner that was much more potent in neonatal animals than in older animals. Consistently, in vivo EEG recordings also revealed that sodium gluconate inhibited the epileptic seizure activity in a manner that was much more potent in neonates than in adult animals. Mechanistically, sodium gluconate inhibits voltage-dependent CLC-3 Cl− channels both in neuronal cultures and in hippocampal slices. Together, these data suggest a novel antiepileptic drug gluconate that potently inhibits neonatal seizures through blocking CLC-3 Cl− channels.

摘要: Computationally efficient accelerated D2-clustering algorithms are disclosed for clustering discrete distributions under the Wasserstein distance with improved scalability. Three first-order methods include subgradient descent method with re-parametrization, alternating direction method of multipliers (ADMM), and a modified version of Bregman ADMM. The effects of the hyper-parameters on robustness, convergence, and speed of optimization are thoroughly examined. A parallel algorithm for the modified Bregman ADMM method is tested in a multi-core environment with adequate scaling efficiency subject to hundreds of CPUs, demonstrating the effectiveness of AD2-clustering.

摘要: Provided are compositions and methods for identifying test agents as candidate for use in reducing hepatitis B virus (HBV). The methods involve a) introducing at least one test agent into mammalian cells, the cells having one or more plasmids that encode an HBV genome, wherein the genome includes a segment encoding a mutated HBV core (HBc), wherein the HBc mutation is L60A, I126A, K96A or a combination thereof. The test agent is allowed to be in contact with the mammalian cells for a period of time. Subsequently, amounts of HBV cccDNA in the mammalian cells are determined. The method can be performed in vitro or in vivo. A reduction in cccDNA relative to a control indicates the test agent in the test container is a candidate for reducing HBV in an individual. Cell cultures divided into reaction containers that each contain a distinct test agent are also included.

摘要: The present disclosure provides a method for killing persister cells with mitomycin C and/or cisplatin, or derivatives thereof. Recalcitrant infections are difficult to treat due to persister cells, a subpopulation of all bacterial populations that is highly tolerant against all traditional antibiotics since the cells are dormant and antibiotics are designed to kill growing cells. Here, we show that MMC and cisplatin eradicate persister cells through a growth-independent mechanism, cross-linking DNA. We find both agents are effective against both planktonic cultures and highly robust biofilm cultures for a broad range of bacterial species, including commensal Escherichia coli K-12 as well as pathogenic species of E. coli, Staphylococcus aureus, and Pseudomonas aeruginosa. In certain approaches cisplatin is superior to MMC.
