    摘要: To detect bands in an electrophoresis capillary tube having a liquid separating medium, a light source and a light detector of a monitor are positioned on opposite sides of the capillary tube with the light source transmitting light through a first slit and light passing to the detector through a second slit after it has passed through the capillary tube. The first and second slits are aligned with the direction of motion of bands, have a maximum length of less than 500 micrometers and a maximum width of less than 200 micrometers. There are no vertical lengths in the capillary tube having a dimension greater than one third of its length between a sample injecting end and a detecting end of the capillary tube. A sample injector includes a vacuum chamber communicating with one end of the capillary tube to pull sample into the other end and a pressure sensor to measure the pressure in said vacuum chamber and generate a signal indicative thereof and integrate it to indicate the amount of sample pulled into the system.

    摘要: To reduce tailing, the frit at the outlet to the liquid chromatographic column has a diameter substantially equal to that of the inside diameter of the outlet end of the column and less than two millimeters. It is held directly against the packing of the column within a cylindrical member having a shoulder with a central aperture at least the size of the diameter of the frit and cylindrical walls which are fastened to resist force in the direction of the axis of the column from its inlet to its outlet. A gasket seals the cylindrical holder against the column wall. The frit is formed of gold-plated nickel spheres sintered together at a temperature lower than the gold-nickel eutectic temperature of 950 degrees Celsius but above the phase-immiscibility temperature of 720 degrees Celsius. The size and shape of the frit is selected to reduce tailing by causing the flow path from the column to be uniform.

    摘要: To stabilize flow rate in a single-stroke, syringe-type high pressure pump to a constant value after turn-on, a control system increases the pump speed until equilibrium conditions are reached and then pumps at the preset rate. In one embodiment, the control system senses the maximum rate of change of pressure, detects a pressure when the rate of change is two-thirds of the maximum, increases the pump rate until the pressure is three times that at the value of the detected pressure and begins pumping at the preset constant flow rate. In other embodiments: (1) a pumping system is operated at a flow rate ten times the preset flow rate and, when the time derivative of the pressure has dropped to nine tenths of its maximum, the pump motor speed is reduced to the preset flow rate; (2) the outflow of the column is measured under no load conditions at the desired flow rate and when a column is connected, the pumping speed is increased until the same outflow results before reducing the pumping rate; and (3) the system is operated in a constant pressure mode to a point at which the pressure is estimated to provide the desired flow rate, the flow rate measured, and if the flow rate is not the preset one, a new pressure measurement is set and the procedure repeated.

    摘要: To scan gels optically, a gel scanner includes a gel holder and an optical system mounted for motion with respect to each other. The optical system includes a tungsten halogen lamp, first aperture, second aperture, beam splitter and first and second light paths; the first aperture having a dimension in the direction of motion of the gel with respect to the optical system of less than 0.5 millimeter and a dimension in a direction transverse to the direction of motion of less than 10 millimeters. The lamp and first aperture are mounted on one side of said gel holder and the second aperture is mounted on the other side of said gel holder. The beam splitter transmits light in a first direction along the first light path and in a second direction along a second light path with the: (1) first light path including a first photodetector and an interference filter for passing light within the range of 400 nanometers wavelength to 700 nanometers wavelength to it; and (2) the second light path having a second photodetector.

In one embodiment, the system is operated in a constant pressure mode under the control of a feedback system to a point at which the pressure is estimated to provide the desired flow rate, with feedback being provided from a pressure measurement and from a flow rate measurement. The feedback from the flow measurement is increased logarithmically with the rate of pumping prior to being compared with the pressure error signal and the comparison between the previous error signal and the logarithmic flow rate signal is used to control the speed of pumping.

In each cassette, the cam is mounted between side plates against which the rollers roll, which are spring biased against the rollers and hinged at a center location to provide precision determined only by the precision of the cam to side plate edge dimension. The cam is shaped to reduce pulses during roller lift-off from the cam surface to provide for pulse-free flow.

    摘要: To reduce noise in an absorbance monitor in which the light source is a feedback controlled deuterium lamp, a mirror focuses light from a part of the central bright spot in the deuterium lamp through a small aperture in an aperture plate which blocks all other light passing through the optical detecting and electrical sensor station. The light passing through the aperture is split into two unpolarized beams within the optical detecting and electrical sensor station, one of which is transmitted through an aperture optically at least as large as the image of the aperture near the lamp and into a flow cell to sense the absorbance or transmittance of an effluent. After the sensing beam has passed through the effluent, it is electrically compared with the other beam which has been passed through an aperture to cancel out the common-mode noise and in some applications the absorbance or transmittance which may be due to the solvent or other carrier of the sample.
