    摘要: Disclosed is a continuous process in which a subset of a number of mutually identical columns, are connected in series. The process liquid, e.g. crude cell culture harvest, is supplied to the most upstream column of the subset. It flows successively through the in series connected columns and leaves the subset through the most downstream and flows into the downstream collection vessel. As soon as the packed bed of the most upstream column is become saturated with product, this column is disconnected from the subset. It is removed from the series connection. A replacement, identical, column is added such that it is connected in series downstream from the most downstream column of the subset. This process is repeated.

    摘要: A durable on-column tunneled frit was developed for use in nanoflow UPLC systems. The frit was tunneled during sol-gel reaction. The tunneled-frit fabrication process is easy and reproducible in terms of back pressure and durability. This design creates low backpressure with high liquid flow, which is suitable for the nanoflowUPLC application. A short packing (2 cm C18 particle) tunneled-frit column was demonstrated to sustain 10,000 psi continuous liquid flow for over one week without any particle leakage or pressure instability. The tunneled-frit was also successfully applied to the fabrication of nanoUPLC trapping and analytical column system. It was demonstrated to have high separation efficiency and sensitivity for the analysis of tryptic peptides as well as improved detection sensitivity for phosphopeptide analysis.

    摘要: A reusable liquid chromatographic column consists of three parts, a column body with an integral outlet port molded on one end, a sealing insert with an inlet port, and a perforated cap providing downward force to compress the sealing insert into the column body. A wedge-like seal is employed to seal the connection between the sealing insert and the column body. Two filters on both ends of the column body are used to prevent escape of packing materials from the column chamber under pressure. Hand tightening is adequate to seal the connection by screwing the perforated cap onto the column body. The column is reusable due to its simple disassembling and reassembling processes. It is also disposable due to its inexpensive fabricating materials and manufacturing process.

    摘要: Described is a reusable fitting for attaching a conduit to a port. The fitting includes a housing, a spring, a retainer and cap. The retainer is capable of locking and unlocking a conduit in the fitting. The fitting provides a high pressure fluid-tight seal and a substantially zero void volume at the junction of two fluidic conduits. The fitting is removable, reusable and replaceable, and allows a user to make and remake connections of a capillary to a fluidic port without permanently binding a ferrule to a specific capillary and thus to a specific port. The fitting can be replaced if part or all of the fitting is damaged during use.
