    摘要: A system and method operative in accordance with the present disclosure facilitate storage and retrieval of individual or discrete samples of biological, non-biological, and chemical material stored on dry media. Sample material may be disposed upon or within a porous or solid (i.e., non-porous) sample storage medium and subsequently archived in, and retrieved from, storage elements such as multi-well plates, for example, using robotic devices or other automated apparatus. The disclosed system and method enable ejection of sample material from a sealed storage element into a specific well of a multi-well daughter plate, or into a specific cuvette, test tube, or similar container. In some embodiments, a sample carrier comprising a storage medium may be punched or ejected through a first seal of the storage element with an apparatus or implement such as a disposable piercing tip, for instance, inserted through a second seal of the storage element.

    摘要: An inner control loop controls the relative position of a surface to be scanned with respect to a focal plane. An outer control loop responds to the location of the surface to be scanned with respect to a focal plane of the scan lens to generate a setpoint for the inner control loop. A sample-and-hold element disposed between the inner and outer control loops is switched by an edge or defect detector. It is preferred that the outer control loop has a time constant in the range of three to ten times longer than that of the inner control loop. The sample-and-hold element provides a gradual state transition upon the occurrence of an edge or a surface defect.

    摘要: A technique for analyzing analytes in a chemical array having a plurality of pixels is provided. The apparatus of the technique contains a light source for irradiating a light beam at the pixels individually, a controller for controlling the relative position of the light source to the array, and a detector for detecting fluorescence resulting from irradiation. The controller controls the light beam generated by the light source to irradiate a first number of pixels sequentially in the array and repeating one or more times the sequential irradiation before irradiating a second number of pixels, the pixels of which are different from those of the first number of pixels. The first number of pixels includes more than one pixel but less than the total number of pixels in the array.

    摘要: A method and system of operating a laser device includes utilizing an integral power-regulation sensor of the laser device to detect energy back reflected from an object of interest and then using the detected signal as a basis for determining a characteristic related to the object of interest. In the preferred embodiment, the determined characteristic is the position of the beam focus relative to a surface of the object. Also in the preferred embodiment, the integral power-regulation sensor is disconnected from power-regulation circuitry during the focusing operation. Interaction of the back reflected energy and the outgoing beam can be reduced by introducing beam rotation, such as by providing a quarter-wave plate along the beam path. Focusing sensitivity can be increased by locating reflective targets over the surface and by executing AC measurements to filter DC components from the signal.
