摘要:
The invention encompasses a novel method of treating inflammatory disease, such as rheumatoid arthritis, and novel methods of identifying and screening for drugs useful in the treatment of inflammatory diseases and their clinical symptoms. The inventors have made the discovery that the activity of HIF-1α, a transcription regulator known to have an effect on some cancers, has a significant impact on the pathophysiology of rheumatoid arthritis. The symptoms of an inflammatory disease, such as rheumatoid arthritis, may be alleviated by administering a compound that inhibits the activity of HIF-1α.

The invention encompasses novel methods of treating rheumatoid arthritis and its symptoms and novel methods of identifying and screening for drugs useful in the treatment of rheumatoid arthritis and its clinical symptoms. Targeted manipulation of a computer model of a human rheumatic joint provided the surprising result that decreasing the activity of FLIP, an inhibitor of apoptosis, by at least 25% has a significant impact on the pathophysiology of rheumatoid arthritis. Inhibition of the activity of FLIP by at least 25% is predicted to alleviate the symptoms of rheumatoid arthritis.

The invention encompasses novel methods of treating rheumatoid arthritis and its symptoms and novel methods of identifying and screening for drugs useful in the treatment of rheumatoid arthritis and its clinical symptoms. Targeted manipulation of a computer model of a human rheumatic joint provided the surprising result that cross-linking soluble Fas-ligand (sFasL) has a significant impact on the pathophysiology of rheumatoid arthritis. The symptoms of rheumatoid arthritis may be alleviated by administering a sFasL-specific cross-linker.

The invention encompasses a novel method of treating an inflammatory disease, such as rheumatoid arthritis, and novel methods of identifying and screening for drugs useful in the treatment of inflammatory diseases, such as rheumatoid arthritis, and their clinical symptoms. The inventors have made the discovery that the activity of galectin-3, a β-galactoside-binding lectin known to have an effect on some cancers, has a significant impact on the pathophysiology of rheumatoid arthritis. The symptoms of an inflammatory disease, such as rheumatoid arthritis, may be alleviated by administering a compound that inhibits the activity of galectin-3.

The invention encompasses novel methods of treating rheumatoid arthritis and its symptoms and novel methods of identifying and screening for drugs useful in the treatment of rheumatoid arthritis and its clinical symptoms. Targeted manipulation of a computer model of a human rheumatic joint provided the surprising result that the activity of CD99, an adhesion molecule known to have an effect on some cancers, has a significant impact on the pathophysiology of rheumatoid arthritis. Inhibition of the activity of CD99 is predicted to alleviate the symptoms of rheumatoid arthritis.

The invention encompasses novel biomarkers and methods for assessing insulin resistance in a subject. The novel biomarkers of the invention include various plasma constituent (e.g., insulin, glucose, lactate and/or triglyceride) concentrations. The methods of the invention include measuring various plasma constituent concentrations and calculating a predicted euglycemic hyperinsulinemic clamp glucose infusion rate (GIR) based on the plasma constituent concentrations.

Methods for predicting effectiveness of a PDE4 inhibitor are described. In one embodiment, a method includes administering a β2-agonist to a patient having asthma and applying a measurement to the patient to determine a treated result of the measurement. The measurement is configured to evaluate effectiveness of the β2-agonist for the patient. The method also includes predicting effectiveness of the PDE4 inhibitor for the patient based on the treated result of the measurement.

The invention encompasses methods, including computer-implemented methods, of defining a virtual patient population and mapping the virtual patient population to a population of real patients. The invention utilizes virtual measures from one or more virtual patients, and data representative of multiple real subjects in a sample population, such as data collected from patients in a clinical trial or epidemiological study of a real population. The invention includes evaluating the similarity between the virtual patients and the real subjects, and assigning prevalences to the virtual patients based on the evaluation. The similarity can be assessed using some or all of the virtual measures of the virtual patients and some or all of the data obtained for the real subjects. Any of various goodness-of-fit measures can be used to evaluate the similarity or to help identify prevalences. The virtual patient population is defined as the virtual patients according to their respective prevalences.

The invention encompasses novel methods for developing a computer model of a mammalian respiratory system. In particular, the models include representations of biological processes associated with obstruction of the respiratory system with constriction of the respiratory system. The invention also encompasses computer models of respiratory systems, methods of simulating respiratory systems and computer systems for simulating respiratory systems.

Apparatus and method for identifying therapeutic targets of a biological system in a disease state are described. In one implementation, a method uses a computer model of the biological system. The method includes identifying a set of functions of a biological constituent of the biological system. The method also includes executing the computer model in the absence of a modification of the set of functions to produce a first output and executing the computer model based on the modification of the set of functions to produce a second output. The method further includes comparing the second output with the first output to identify the biological constituent as a therapeutic target.
