摘要: The present disclosure relates to methods for detecting functional autoantibodies and to diagnostic and prognostic applications of the methods. In certain embodiments, the present disclosure provides a method of detecting a functional autoantibody against an antigenic target, the method comprising exposing a cell expressing the antigenic target to the autoantibody, wherein the antigenic target has an activity to alter the level of reactive oxygen or nitrogen species in the cell and binding of the autoantibody to the antigenic target alters the activity of the antigenic target, and detecting the functional antibody on the basis of a change in the level of the reactive oxygen or nitrogen species in the cell.

摘要: A method of identifying a detectable molecular signature for an antibody or antibodies of interest (eg autoantibodies associated with one or more autoimmune diseases) wherein the molecular signature comprises an Unusual or common heavy (H)-chain and light (L)-chain pairing and/or at least one mutation in one or more of said H- and L-chains and/or an adjacent region. The detection of antibodies comprising such a molecular signature, preferably by using a targeted mass spectroscopy (MS) approach to directly sequence the antibodies or a part(s) thereof, can provide a diagnostic biomarker for one or more disease or disorder.

摘要: The present invention relates generally to the field of tissue modification, and in particular, but not exclusively, to methods of modifying cells of ocular and particularly corneal tissue to produce immunoglobulins or immunoglobulin fragments. The invention also relates to modified ocular cells and tissues, to expression vectors utilised in such methods, to methods of xeno- and allo-transplantion using the modified tissue and to methods of therapy of ocular diseases and disorders. In a preferred embodiment of the invention there is provided a method of modifying cells of ocular tissue to produce an immunoglobulin or immunoglobulin fragment of interest, which comprises exposing ocular tissue to an effective concentration for transfection of an expression vector comprising a nucleotide sequence encoding for the immunoglobulin or immunoglobulin fragment, for a period sufficient to allow transfection, such that cells of said ocular tissue produce the immunoglobulin or immunoglobulin fragment.

摘要: This invention relates to a method for the isolation of trophoblast (placental) cells from the blood of a pregnant mammal so as to provide the essential starting material, namely cells derived from the fetus, to enable genetic and/or biochemical information about the fetus to be obtained. In particular, this invention relates to the use of monoclonal antibodies specific for membrane protein markers on mammalian trophoblasts to isolate trophoblast cells from maternal blood. These cells may then be used to obtain fetal genetic and/or biochemical information early in pregnancy. The present invention is particularly relevant for detecting human fetal abnormalities.

摘要: A method of enhancing embryo implantation in a subject is disclosed which comprises administering to the uterine cavity of the subject a formulation comprising copper and/or zinc in an amount effective to stimulate endometrial production of leukaemia inhibitory factor (LTF) and' or vascular endothelial growth factor ( VEGF). Alternatively, a device may be inserted into the uterine cavity of the subject, wherein the device comprises copper and/or zinc, for a period of time that is effective to stimulate endometrial production of LIF and/or VEGF. The method is suitable for use with women undergoing treatment by any of the assisted reproductive technologies, such as those involving the transfer of embryos such as in vitro fertilisation (IVF) and variants including IVF-ICSI (intracytoplasmic spenn injection) and m vitro maturation (1VM) treatments, as well as intrauterine-insemination (IUI) therapy. However, the method is also applicable for women wanting to improve their prospects of pregnancy through natural conception.
