In various aspects, the invention provides methods for cyclizing proteins, including methods for enhancing the stability of cyclized proteins under cytosolic conditions. The invention also provides various methods for using the cyclized proteins. For example, cyclized proteins of the invention may be used in screening assays analogous to the yeast two hybrid assay. Selected embodiments of the invention provide cyclized single chain variable fragment (ScFv) molecules, including molecules in the form of an immunoglobulin fold.

The present invention relates to 4-piperidone derivatives represented by the following formula (I) and the acid addition salts thereof. The method of preparation and antineoplastic activity of the said compounds are disclosed. A number of the compounds possess submicromolar IC50 and CC50 values and have a selective toxicity for colon cancers and leukemic cells. In addition, many of the compounds are able to reverse multidrug resistance.

Detection of elevated levels of N-myristoyltransferase (NMT) or NMT activity in blood or bone marrow, and specifically in peripheral blood mononuclear cells, can be used as a marker for cancer. The use of this method for detection of adenocarcinoma, such as colorectal cancer, is exemplified.

Also described is the use of the GapC proteins from those species in vaccine compositions to prevent or treat bacterial infections in general, and mastitis in particular.

The invention provides a delivery system for a biologically active agent comprising a gemini surfactant in admixture with the biologically active agent in a topical formulation for treatment of skin disorders or metabolic diseases. The invention also provides pharmaceutical compositions in topical formulations comprising the delivery system as set out above, in admixture with pharmaceutically acceptable carriers, diluents, excipients, or supplements suitable for application to the skin or mucosal membrane. Methods of preparation and use are also described.
