Various bispecific antibodies that specifically bind to both blood coagulation factor IX/activated blood coagulation factor IX and blood coagulation factor X and functionally substitute for the cofactor function of blood coagulation factor VIII, that is, the function to promote activation of blood coagulation factor X by activated blood coagulation factor IX, were produced. From these antibodies, multispecific antigen-binding molecules having a high activity of functionally substituting for blood coagulation factor VIII were successfully discovered.

Giving attention to the fact that no secretion occurs by H chain in one side due to "knobs-into-holes", it is found out that a target bispecific antibody can be preferentially formed by first expressing H chain and L chain in one side, after inhibiting the expression, expressing H chain and L chain in the other side to construct target HL molecules (HaLa and HbLb) and then pairing the H chains together (H2L2).

A method of screening a common light chain which comprises: (a) the step of producing a host secreting the heavy chain of an antibody binding to a desired antigen; (b) the step of transferring an antibody light chain library into the host of the step (a) and thus producing libraries presenting antibodies consisting of the above heavy chain and the above light chain; (c) selecting a library presenting an antibody binding specifically to the desired antigen as described in the step (a); (d) the step of transferring the library selected in the step (c) into a host secreting the heavy chain of an antibody binding to a desired antigen, which is different from the antigen of the step (a), and thus producing libraries presenting antibodies consisting of the heavy chain and the light chain; and (e) selecting a library presenting an antibody binding specifically to the desired antigen as described in the step (d).

The present inventors produced a variety of bispecific antibodies that specifically bind to both F. IX/F. IXa and F. X, and functionally substitute for F. VIIIa, i.e., have a cofactor function to promote F. X activation via F. IXa. Among these antibodies, the antibody A44/B26 reduced coagulation time by 50 seconds or more as compared to that observed when the antibody was not added. The present inventors produced a commonly shared L chain antibody from this antibody using L chains of A44, and showed that A44L can be used as commonly shared L chains, although the activity of the resulting antibody is reduced compared to the original antibody (A44HL-B26HL). Further, with appropriate CDR shuffling, the present inventors successfully produced highly active multispecific antibodies that functionally substitute for coagulation factor VIII.

An agonistic antibody can be efficiently screened by superinfecting a cell strain, which proliferates depending on a ligand (factor), with antibody libraries respectively to various receptor chains and collecting an antibody gene from an autonomously growing strain by autocline of an agonistic antibody formed from an appropriate combination. Since antibodies are used as libraries in this method, the screening can be performed efficiently and conveniently without resort to troublesome operations.
