Abstract: Embodiments disclose a device for testing biological specimen. The device includes a receiving mechanism to receive a carrier. The carrier includes a holding area that carries or has been exposed to the biological specimen. The device includes a camera module arranged to capture imagery of the holding area. The camera module includes an focusing motor operable to adjust a focal point of the camera. The device also includes a processor that is configured to utilize the camera module to determine, based on operations of the focusing motor, a volumetric property of the holding area and perform a set of analytic processes on at least a portion of the captured imagery of the holding area to determine one or more properties of the biological specimen.

Abstract: Embodiments disclose a device for testing biological specimen. The device includes a sample carrier and a detachable cover. The sample carrier includes a specimen holding area. The detachable cover is placed on top of the specimen holding area. The detachable cover includes a magnifying component configured to align with the specimen holding area. The focal length of the magnifying component is from 0.1 mm to 8.5 mm. The magnifying component has a linear magnification ratio of at least 1. Some embodiments further include a multi-camera configuration. These embodiments include a first camera module and a second camera module arranged to capture one or more images of the first holding area and the second holding area, respectively. The processor may perform different analytic processes on the captured images of different holding areas to determine an outcome with regard to the biological specimen.

Abstract: Disclosed herein are methods for the detection of the presence of sperm DNA fragmentation in a semen sample. The methods include embedding of sperm cells of the semen sample in a gel, denaturing DNA of the sperm cells, and lysing the nuclear proteins of the sperm cells. The present method includes an ionic surfactant sodium dodycyl sulfate (SDS) and a chaotropic agent urea in the lysis solution for releasing DNA from protamine of chromosome, which significantly reduces the time required for lysis. A kit for detecting sperm DNA fragmentation in a semen sample is also disclosed.

Abstract: Disclosed are verification techniques that can be implemented by a device that conducts biological tests to verify that the specimen holding area carries a valid biological specimen. In certain embodiments, the testing device includes a receiving mechanism to receive a carrier, and the carrier includes a holding area that is to carry or to be exposed to a biological specimen. The device can also include a camera module arranged to capture imagery of the carrier, and a processor. In some examples, the processor can capture the imagery of the carrier and identify a visual cue on the carrier. Then, the processor can verify, based on a manner of how the visual cue is displayed in the captured imagery, whether the holding area carries a valid biological specimen.

Abstract: Disclosed are calibration techniques that can be implemented by a device that conducts biological tests. In certain embodiments, the device for testing a biological specimen includes a receiving mechanism to receive a carrier, a camera module arranged to capture imagery of the carrier, and a processor. Some examples of the processor can detect a calibration mode trigger. In calibration mode, the processor can divide the captured imagery into segments and selectively perform one or more calibration procedures for each segment. Then, the processor records a calibration result for each segment.

The device may include a positioning mechanism operable to adjust a relative location of the carrier to the camera. A processor in the device may utilize the camera module to: identify an edge of the first image; cause the positioning mechanism to adjust the relative location of the carrier to the camera in a manner such that, when the camera takes the second image, an edge of the second image aligns with the identified edge of the first image; and perform a set of analytic processes on a combined image from the first and second images.
