Abstract:
The invention provides an improved method for the purification of nucleic acid molecules, which method comprises generating a cellular lysate containing the nucleic acid; contacting the lysate with an anion exchanger bound to a solid support matrix under conditions such that the anion exchanger binds the nucleic acid; followed by eluting the nucleic acid from the anion exchanger with an aqueous mobile phase comprising an elution solution; and desalting the eluted nucleic acid such that it is suitable for downstream applications. The improvement of the method includes adding in the elution solution a composition such that the pH of the aqueous mobile phase is between about pH 9 and about pH 13, wherein the presence of the composition in the elution solution provides an increase in nucleic acid recovery, as compared with the recovery in the absence of the composition.

A multiple-beam antenna system includes at least one reflector and a cluster of horns for feeding the reflector. A horn of the cluster of horns is configured for providing transmission and reception of signals over respective transmission and reception frequency bands. The horn includes a substantially conical wall having an internal surface with a variable slope. The internal surface of the substantially conical wall includes slope discontinuities. At least one of the slope discontinuities has a diameter greater than 1.7 times the wavelength of the lowest frequency of the transmission frequency band. The diameter is also greater than 1.7 times the wavelength of the highest frequency of the transmission frequency band. In addition, the diameter is greater than 1.7 times the wavelength of the lowest frequency of the reception frequency band, and the diameter is greater than 1.7 times the wavelength of the highest frequency of the reception frequency band. This configuration of the slope discontinuity generates one or more higher order modes of a transverse electric (TE) mode over the transmission and reception frequency bands without generating a transverse magnetic (TM) mode.

Disclosed is a process for separating and/or purifying a nucleic acid by elution of the nucleic acid from anion exchange resins under conditions of high salt concentration and the presence in the eluant of an additive comprising guanidine, or a guanidine-like derivative. The process allows high recovery of nucleic acids from anion exchange resins without impairing the nucleic acid stability as compared with conventional ion exchange chromatographic procedures.

An antenna system includes a reflector having a modified-paraboloid shape; and a multi-beam, multi-band feed array located at a focal point of the reflector so that the antenna system forms a multiple congruent beams that are contiguous. The system has a single reflector with non-frequency selective surface. The reflector is sized to produce a required beam size at K-band frequencies and is oversized at EHF-band frequencies. The synthesized reflector surface is moderately shaped and disproportionately broadens EHF-band and Ka-band beams compared to K-band beams. The synthesized reflector surface forms multiple beams each having a 0.5-degree diameter at K-band, Ka-band, and EHF band. The multi-beam, multi-band feed array includes a number of high-efficiency, multi-mode circular horns that operate in focused mode at K-band and defocused mode at Ka-band and EHF-band by employing “frequency-dependent” design for the horns.

Exploitation of suitably functionalized heterocyclic molecules, in the design and synthesis of Fluorescence Resonance Energy Transfer (FRET) cassettes and their corresponding dideoxynucleotide terminators culminated into efficient reagents for DNA sequencing. Additionally, these FRET cassettes/terminators, of the present invention, derived from different classes of heterocyclic systems have high potential to be used for general labelling of biological molecules to generate highly sensitized signals. Their preparation, energy transfer efficiency, and use as labels, specifically, in DNA sequencing reactions is disclosed.

Lonidamine derivatives can be useful in methods of treating, inhibiting, and/or preventing polycystic kidney disease (PKD). Accordingly, lonidamine derivatives can be administered in a therapeutically effective amount for inhibiting, and/or preventing polycystic kidney disease (PKD) in the subject. This can include administering a therapeutically effective amount of the lonidamine derivatives for inhibiting CFTR and/or Hsp90 or biological pathway thereof. Also, the method can include administering the lonidamine derivatives in a therapeutically effective amount for inhibiting ErbB2, Src, Raf-1, B-Raf, MEK, Cdk4, NKCC1, or combinations thereof. For example, the therapeutically effective amount of the lonidamine derivatives can be configured so as to provide a concentration in or adjacent to a kidney cell of about 0.25 uM or more or less.

Fertility management can also include administering an effective amount of the compound to: impair Sertoli cell function in a male subject; inhibit spermatogenesis in the subject; reduce testis weight in the subject; reduce ovary weight in a female subject; reduce serum progesterone in the female subject; impair ovarian follicle function in the female subject; causing reversible fertility in the subject. In order to return fertility, the method can include ceasing administration of the compound to the subject so as to return fertility in the subject. The compound can be administered for irreversibly sterilizing the subject.

The method includes obtaining input data representative of a first parameter of a cryogenic medium in a cryogenic cooling system is provided. The method further includes calculating an expected value representative of a second parameter of the cryogenic medium in the cooling system via a predefined physics based model. A second input data representative of the second parameter of the cryogenic medium in the cooling system is estimated. The expected value is compared to the second input data and difference between the expected value and the second input data is correlated to statistical data to estimate a likelihood of cryogenic cooling system event.
