Abstract:
A method of detecting a molecule associated with viability of one or more cells or organisms in a sample comprises the initial step of contacting the sample with an enzyme, which enzyme is capable of adding or removing a chemical moiety to or from a nucleic acid molecule in the presence of the molecule associated with viability of the of the one or more cells or organisms. This thereby generates a novel detectable nucleic acid molecule. The next step involves detecting the presence of the molecule associated with viability of the one or more cells or organisms by detecting the novel nucleic acid molecule generated only in the presence of the molecule associated with viability of the one or more cells or organisms. A most preferred molecule associated with viability is ATP, although NAD may also be detected. A preferred enzyme for use in the methods is ligase. The method has numerous applications, in particular in monitoring viability of cells, toxicology testing and determining whether there is contamination in a sample or on a surface. Kits are also provided for carrying out the methods.

An improved method of detecting an enzyme in a sample, which enzyme is capable of adding or removing a chemical moiety to or from a nucleic acid molecule, thereby conferring the nucleic acid molecule with the ability to be extended to generate a novel detectable nucleic acid molecule, comprises the steps of allowing the sample to be tested for the presence of the enzyme to interact with the nucleic acid molecule; and testing for interaction of the enzyme with the nucleic acid molecule by detecting the novel nucleic acid molecule generated only in the presence of the enzyme. The preferred enzyme is a phosphatase. The methods have a number of applications, for example in enhancing the sensitivity of immunoassays, for detecting pathogen associated phosphatase, for diagnosing certain conditions and for detecting specific contaminants in a sample.

A microelectronic package includes a microelectronic element having faces and contacts and a flexible substrate spaced from and overlying a first face of the microelectronic element. The package also includes a plurality of conductive posts extending from the flexible substrate and projecting away from the first face of the microelectronic element, wherein at least some of the conductive posts are electrically interconnected with the microelectronic element, and a plurality of support elements supporting the flexible substrate over the microelectronic element. The conductive posts are offset from the support elements to facilitate flexure of the substrate and movement of the posts relative to the microelectronic element.

A structural beam has a thin metal web with a plurality of holes located along its top and bottom edges. One or more rectangular flange pieces are located on each side of the web overlying its top margin and one or more rectangular flange pieces are located on each side of the web overlying its bottom margin. The flange pieces contain holes which are aligned with the holes in the web piece and fasteners extend through the aligned holes to fasten the flange pieces to the web.

The assembly has two lamps, such as fluorescent tubes. A cover is a co-extrusion having a baffle between the lamps and shroud portions outermost of the cover. The baffle blocks at least some of the light output of the lamps from crossing the region between the lamps. This helps reduce glare, which is further reduced by the configuration of the shrouds.

A can opener has a first and a second handle member pivotably connected to be moveable between an open disposition and a closed disposition. They can be squeezed together in one hand of the operator to rotate a drive wheel. A cutting wheel is mounted onto the first handle member. A radially facing aperture is formed through a part of said first handle member so that the drive wheel may partly protrude to engage the rim of the can and press it against the cutting wheel. The drive wheel is mounted rotatably onto a rivet which is held slideably in a slot in the main plate of the first handle member to be moveable towards and away from the cutting wheel.

An airfoil blade, such as a jet engine turbine rotor blade. An internal serpentine coolant circuit has a last downstream passageway bounded by four monolithic inner walls which are monolithic with at least a portion of the outer walls. Two of the inner walls are spaced from the outer walls and contain air impingement orifices creating two impingement chambers. Some coolant in the serpentine circuit exits the airfoil blade through a coolant exit in the blade tip. The remaining coolant in the circuit passes through the impingement orifices and exits the blade through film cooling holes in the outer walls.

A jet engine component, such as an aircraft gas turbine engine rotor blade or a scramjet engine fuel injector. The component has a wall portion including a first surface exposable to a cooler, higher static pressure fluid and a second surface exposable to a hotter, lower static pressure gas flow flowing across the second surface. The component further includes a generally straight film coolant passageway having an inlet on the first surface and an outlet on the second surface. The second surface has a seamless groove which is open substantially entirely along its longitudinal dimension extending from the outlet along the lower static pressure gas flow for improved film cooling of the second surface.
