a method is provided for detecting a presence of HIV virus in a sample comprising: taking a culture of recombinant cells which (a) are capable of cell division, (b) express CD4 receptor and one or more additional cell surface receptors necessary to allow the HIV virus to infect, (c) enable the HIV virus to replicate and infect the noninfected cells in the cell culture, and (d) comprise a reporter sequence introduced into the recombinant cells comprising a reporter gene whose expression is regulated by a protein specific to HIV viruses which is expressed from a genome of an HIV virus upon infection of the recombinant cell by the HIV virus; contacting the cell culture with a sample to be analyzed for the presence of HIV virus in the sample; and detecting a change in a level of expression of the reporter gene in cells in the recombinant cell culture. The method can be used to detect the presence of HIV virus in a sample, detect the presence of different strains of HIV virus in a sample, detect HIV drug resistance in a sample, determine what combination of one or more anti-HIV agents would be effective in treating a patient, and screen compositions for anti-HIV activity.

Immortalized mammalian dendritic cell lines are provided that have characteristics of functional, mature dendritic cells; defined by surface phenotype, constitutive and inducible gene expression patterns, and functional effects on T lymphocytes. The cell lines are capable of continuous in vitro proliferation for extended periods of time. Of particular interest is the ability of the cell lines to present antigen in a stimulatory manner to naive T cells. The dendritic cell lines are useful in gene discovery, the generation of dendritic cell subset-specific antibodies and probes; defining stages of dendritic cell differentiation and maturation; for induction of antigen specific stimulation of lymphocytes; for dissection of antigen specific anergy or tolerance in lymphocytes; for characterization of dendritic cell-natural killer cell interactions; for in vitro and in vivo analyses of dendritic cell-endothelial cell interactions; and for screening assays.

DNA encoding such proteins in either the sense or anti-sense orientation can be administered to either enhance or inhibit the cellular response to IFNs. Antibodies to the proteins can be used for isolation of the new protein or for immunodetection thereof.

The present invention relates to a novel T1-R ligand III protein which is a member of the IL-1RI ligand-like family. In particular, isolated nucleic acid molecules are provided encoding the human T1-R ligand III protein. T1-R ligand III polypeptides are also provided as are vectors, host cells and recombinant methods for producing the same. The invention further relates to screening methods for identifying agonists and antagonists of T1-R ligand III activity. Also provided are diagnostic methods for detecting immune system-related disorders and therapeutic methods for treating immune system-related disorders.
