摘要: The inventors have developed a method of modifying the amino acid sequence of a CGTase to obtain variants. The variants may form linear oligosaccharides as an initial product by starch hydrolysis and a reduced amount of cyclodextrin and may be useful for anti-staling in baked products. The method is based on a comparison of three-dimensional (3D) structures of the CGTase with the structure of a maltogenic alpha-amylase where one or both models includes a substrate. The invention also provides novel CGTase variants.

摘要: The present invention relates to variants of a parent α-amylase, which parent α-amylase (i) has an amino acid sequence selected from the amino acid sequences shown in SEQ ID No. 1, SEQ ID No. 2, SEQ ID No. 3, and SEQ ID No. 7, respectively; or (ii) displays at least 80% homology with one or more of these amino acid sequences; and/or displays immunological cross-reactivity with an antibody raised against an α-amylase having one of these amino acid sequences; and/or is encoded by a DNA sequence which hybridizes with the same probe as a DNA sequence encoding an α-amylase having one of these amino acid sequences; in which variant:(a) at least one amino acid residue of the parent α-amylase has been deleted; and/or (b) at least one amino acid residue of the parent α-amylase has been replaced by a different amino acid residue; and/or (c) at least one amino acid residue has been inserted relative to the parent α-amylase; the variant having α-amylase activity and exhibiting at least one of the following properties relative to the parent α-amylase: increased thermostability; increased stability towards oxidation; and reduced Ca2+ dependency; with the proviso that the amino acid sequence of the variant is not identical to any of the amino acid sequences shown in SEQ ID No. 1, SEQ ID No. 2, SEQ ID No. 3 and SEQ ID No. 7, respectively.

摘要: The pharmaceutical use of amylases related to Bacillus alpha-amylases of SEQ ID NOs: 1-3, optionally in combination with a lipase and/or a protease. Examples of medical indications are: Treatment of digestive disorders, pancreatic exocrine insufficiency (PEI), pancreatitis, cystic fibrosis, diabetes type I, and/or diabetes type II. The amylases of SEQ ID NOs: 1-3 are variants of amylases from Bacillus stearothermophilus, Bacillus licheniformis and Bacillus sp. The amylases of the invention have an improved efficacy in vivo, an improved pH-profile, a high specific activity, and/or an improved starch degradation profile.

摘要: The enantioselectivity of fungal lipolytic enzymes can be altered by substituting a suitably selected amino acid residue. The residue to be substituted is selected from its location in the 3D structure of the enzyme and an ester substrate (or a substrate analogue). A residue in the lid may be selected if it is located close to the acid part or close to the alcohol part of an ester substrate. A residue outside the lid region may be selected if it is located close to the active site or close to the substrate.
