Various methods for controlling one or more parameters of a flow cytometer type measurement system are provided. One embodiment includes monitoring parameter(s) of the measurement system during measurements of sample microspheres. The method also includes altering the parameter(s) in real time based on the monitoring. Another method includes monitoring a temperature proximate to the measurement system. One such method includes altering a bias voltage of an avalanche photo diode in response to the temperature using empirically derived data. A different such method includes altering output signals of a photomultiplier tube in response to the temperature using a characteristic curve. Some methods include monitoring a temperature of a fluid, in which sample microspheres are disposed, that will flow through the flow cytometer type measurement system. This method also includes determining a velocity of the sample microspheres in the measurement system from a viscosity of the fluid at the temperature.

A particle generating system includes an aerosol generator, an ejector diluter, and an aerosol diluter. The ejector diluter receives the generated aerosol and dilutes the aerosol to an expected raw concentration. The aerosol diluter further dilutes the aerosol to a concentration in the range of 0% to 100% of the expected raw concentration. The aerosol diluter includes a mini cyclone for diluting the aerosol. The particle generating system may be configured to provide variable concentrations of monodisperse or polydisperse aerosols for instrument calibration. The system may provide constant concentrations in the range of 0% to 100% of the raw concentration. The mini cyclone makes the system compact, and the system may be portable.

A method for the simultaneous determination of the total concentration of sperm cells and the proportion of live sperm cells in a semen sample is provided. By selective staining of live, dead and/or dying sperm cells, for example using one or more fluorochromes, the detection means can distinguish between live, dead and/or dying sperm cells, for example by detection of the fluorescent response from the sperm cells. The selective staining may be performed at room temperature and in concentrations below standard concentrations. The determination may be performed by selective counting of cells of each fluorescent quality for example by using a flow cytometer having internal concentration standard means. Furthermore, the likelihood of fertilizing a female animal with an insemination dose, taken from a semen sample having a known total concentration of sperm cells and a known proportion of live sperm cells, is predicted on the basis of the thus determined total concentration of sperm cells and the proportion of live sperm cells.

Procedure for controlling the enumeration of the absolute count of cells (or other particles) present in a sample. The procedure consists of the following: to prepare, in known quantities and proportions, a mixture or stock solution of two or more populations of reference particles of differing characteristics; to add a known quantity of this mixture of reference particles to a known volume of the sample which contains the cells (or other particles) to be counted; to measure, in a flow cytometer, the sample containing both the cells (or other particles), the events of which are to be counted, and the mixture of different populations of reference particles; to calculate the absolute number of cells (or other particles) present in the sample to be counted and; to check that the proportion between the different reference particles present in the sample measured concurs with the proportion that exists between them in the initial mixture or stock solution which contained the different populations of reference particles prior to adding it to the sample.
