    Abstract: The specificity of binding of a zinc finger to a triplet or quadruplet nucleotide target subsite depends upon the location of the zinc finger in a multifinger protein and, hence, upon the location of its target subsite within a larger target sequence. The present disclosure provides zinc finger amino acid sequences for recognition of triplet target subsites having the nucleotide G in the 5′-most position of the subsite, that have been optimized with respect to the location of the subsite within the target site. Accordingly, the disclosure provides finger position-specific amino acid sequences for the recognition of GNN target subsites. This allows the construction of multi-finger zinc finger proteins with improved affinity and specificity for their target sequences, as well as enhanced biological activity.

    Abstract: The present disclosure provides methods for producing acetaldehyde from renewable biological resources, for example, from a fermentable substrate, with the advantages of energy efficiency and ease of purification. Particular embodiments, feature the production of acetaldehyde from pyruvate, wherein the pyruvate is generated from various carbon sources (e.g., sugars) by pyruvate-producing microorganisms. The methods comprise culturing a pyruvate-producing microorganism in a culture medium under conditions such that pyruvate is excreted and accumulates extracellularly, resulting in a pyruvate-enriched medium.

    Abstract: Modified forms of naturally occurring bacteriocins, such as the R-type pyocins of Pseudomonas aeruginosa, are disclosed. The bacteriocins are modified at the ends of their tail fibers in a region responsible for binding specificity and affinity to their cognate binding partners, or receptors, such as those on the surface of bacteria. Methods for the use of the modified bacteriocins, such as to bind receptors, including virulence or fitness factors, on the surfaces of bacteria, are also described.
