Methods of diagnosing Clostridium difficile infection and compounds used in the methods are provided. The diagnostic methods are specific and sensitive methods for detecting a host cell response and, in some aspects, a target response, i.e. a Clostridium difficile toxin, in an individual infected with Clostridium difficile. The methods comprise detecting in a stool specimen or fluid exposed to the stool specimen a Clostridium difficile toxin, or a fragment thereof, and an increase in a colonic epithelial cell protein exemplified by a non-muscle tropomyosin. Also provided are kits comprising reagents for detecting host cell proteins and Clostridium difficile toxins.

A system and method for transferring component panes from a component pane storage station to a vacuum table assembly are disclosed. The system includes a number of component pane handlers, for instance two handlers, and a pick and place mechanism or arm. The handler transfers the component panes from the station to the mechanism. The system includes an alignment module configured to facilitate spatial alignment of the component pane while the component pane is coupled to the handler. The module can be carried by the mechanism. Spatial alignment of the component pane can be substantially completed before transfer of the component pane from the handler to the mechanism. The mechanism includes a set of vacuum elements or pads that enables pick up of the component pane from the handler as well as release and pick up of the component pane at the vacuum table assembly.

The displacement of the component pane capture element can be effectuated by a displacement of at least a portion of the displacement arm. A method for handling component panes of multiple sizes is also included in this disclosure.

The disclosure provides specific and sensitive anti-toxin B antibodies and fragments thereof suitable for diagnosing Clostridium difficile infection. The antibodies and fragments recognize an epitope in the C-terminal 250-amino-acid region of toxin B of C. difficile, including epitopes defined by protein repeat sequences in this region of toxin B. This disclosure also provides the toxin B-specific epitope in the C-terminal 250-amino-acid region of toxin B of C. difficile for use in vaccine development as well as in the treatment of CDI disease and in treatment of the relapse of CDI disease. Also provided are toxin B polypeptides lacking the cytotoxic domain useful in treating or preventing CDI disease. PCR-based diagnostic assays targeting the 750-nucleotide region at the 3′ end of tcdB are also provided.
