Methods are described for performing a multiplexed analysis of a level of target analyte in a sample, employing an identifier and a labeling reagent. Either or both of the identifier and the labeling reagent comprises a SERS-active nanoparticle associated with a SERS-active reporter with a uniquely identifiable spectroscopic signature. Interrogation of the identifier and the labeling reagent is conducted by serial coincident detection. Such methods can provide enhanced multiplexed analysis of analytes in a sample, especially with regards to improving the type of identifying reagents that are employed.

A lateral flow device is disclosed. The lateral flow device includes a substrate having a flow path and a detection zone disposed along the flow path. The detection zone includes an immobilized target-binding moiety directed against a target of a Raman-active complex. Also disclosed is a method of conducting a lateral flow assay and detection system. The method includes i) defining a flow path having a detection zone; ii) flowing a sample down the flow path; and iii) immobilizing a Raman-active complex if present, at the detection zone. The sample includes a Raman-active complex or a Raman-active tag.

Disclosed herein are biosensors and methods for making and using the same. In one embodiment, the sensor for detecting an analyte comprises: a substrate, recognition elements specific for the analyte, an excitation source, a detector, a chamber located between the substrate and the excitation source and between the substrate and the detector, and an emission filter. The recognition elements are tethered to the substrate such that the recognition elements can be exposed to a sample. The excitation source is capable of emitting a first light having a first light peak intensity at a first wavelength, wherein the first light can excite a luminophore to emit a second light when the recognition elements interact with the analyte. The detector is capable of detecting the second light emitted by the luminophore. The emission filter is capable of filtering in a band gap that includes the first light peak intensity.

Other embodiments an RFID tag and at least one antibody coupled to the RFID tag. The RFID tag includes a substrate, circuitry disposed on the substrate, and an antenna coupled to the substrate. The at least one antibody is capable of affecting the signals emanating from the RFID tag. Further embodiments include a detection system that includes a reader. Yet other embodiments include a method for detecting specific analytes. The method includes providing an RFID tag which emanates a first signal having a first frequency, and enabling the REID tag to emanate a second signal having a second frequency upon attraction of a specific analyte to the RFID tag.

The process generally includes the competitive or non-competitive binding of target substances onto carrier particles functioning as substrates in the biological assay. Employing the carrier particles as substrates provides increased surface area for the reaction to occur; increased ease of washing steps; and allows for concentration of the increased surface area into a smaller reaction volume prior to introduction into the vapor and/or gas phase spectrometer such as an ion mobility spectrometer.
