    摘要: An apparatus and method are provided for creating an image of a microarray. The apparatus includes at least one light source configured to direct light toward the microarray. The apparatus includes an excitation filter configured to filter the light into a first frequency band towards dichromatic mirror. The dichromatic mirror reflects light onto the microarray causing the microarray to emit electromagnetic energy. The apparatus includes emission filter configured to filter the electromagnetic energy within a second frequency band. The apparatus further includes an imaging unit having a charged coupled device (CCD), the CCD having an imaging surface masked by a pinhole blind such that when the pinhole blind receives electromagnetic energy from the emission filter, an image is created of the entire micro array.

    摘要: An apparatus and method are provided for creating an image of a microarray. The apparatus includes at least one light source configured to direct light toward the microarray. As raw image data is received from a sequencing machine, it is buffered and then separately transformed in real time in the processors to enhance the signals indicative of the unknown DNA sequence. A fourth processor receives the transformed data and determines and reports the sequence indicating events.

    摘要: A method and system for automated nucleic acid sequence determination of a polynucleotide, wherein a nucleic acid sequencing ladder comprises signals corresponding to oligonucleotides formed from the polynucleotide, comprising the step of correlating, particularly in a trained neural network or a scatter plot, an intensity variable for each signal in the nucleic acid sequencing ladder with an informative variable for that signal, wherein the informative variable comprises information from at least two adjacent signals in the nucleic acid sequencing ladder, such that each signal in the nucleic acid sequencing ladder identified so as to determine the nucleic acid sequence corresponding to the polynucleotide. In particular, the relative separation between consecutive signals, the relative intensities between consecutive signals, and a pattern recognition factor, which incorporates a comparison of relative separations and intensities of at least two adjacent signals with pattern recognition templates, can be used as informative variables. Furthermore, this invention relates to a method and system for the on-the-fly resolution and extraction of information of signals contained in a digitized data stream involving calculation of the smoothed second derivative of a data point from the smoothed first derivative of the data point.
