摘要: A microfluidic device for determining whether an analyte is present in a sample is provided. The microfluidic device includes an elongated flow path having a polymeric medium, where the polymeric medium includes a first analyte detection domain having a first immobilized capture member that specifically binds to a first analyte and a second analyte detection domain having a second immobilized capture member that specifically binds to a second analyte. Also provided are methods, systems and kits in which the subject microfluidic devices find use, as well as methods of producing the same.

摘要: Provided is a gene analysis method including performing a single-base extension reaction using a primer for a single-base extension reaction for detection of a target nucleotide sequence, and a substrate for a single-base extension reaction having a fluorescent dye; subjecting a reaction product of the single-base extension reaction to electrophoresis; and measuring mobility of the electrophoresis and fluorescence intensity of the fluorescent dye, and detecting a wild type and a mutant of the target nucleotide sequence based on the fluorescence intensity. Also provided is a kit for use in the gene analysis method disclosed herein.

摘要: Apparatus and techniques for electrokinetic loading of samples of interest into sub-micron-scale reaction chambers are described. Embodiments include an integrated device and related apparatus for analyzing samples in parallel. The integrated device may include at least one reaction chamber formed through a surface of the integrated device and configured to receive a sample of interest, such as a molecule of nucleic acid. The integrated device may further include electrodes patterned adjacent to the reaction chamber that produce one or more electric fields that assist loading the sample into the reaction chamber. The apparatus may further include a sample reservoir having a fluid seal with the surface of the integrated device and configured to hold a suspension containing the samples.

摘要: The invention provides a passive fluidics circuit for directing different fluids to a common volume, such as a reaction chamber or flow cell, without intermixing or cross contamination. The direction and rate of flow through junctions, nodes and passages of the fluidics circuit are controlled by the states of upstream valves (e.g. opened or closed), differential fluid pressures at circuit inlets or upstream reservoirs, flow path resistances, and the like. Free diffusion or leakage of fluids from unselected inlets into the common outlet or other inlets at junctions or nodes is prevented by the flow of the selected inlet fluid, a portion of which sweeps by the inlets of unselected fluids and exits the fluidics circuit by waste ports, thereby creating a barrier against undesired intermixing with the outlet flow through leakage or diffusion.

摘要: Techniques described herein can apply AC signals with different phases to different groups of nanopore cells in a nanopore sensor chip. When a first group of nanopore cells is in a dark period and is not sampled or minimally sampled by an analog-to-digital converter (ADC) to capture useful data, a second group of nanopore cells is in a bright period during which output signals from the second group of nanopore cells are sampled by the analog-to-digital converter. The reference level setting of the ADC is dynamically changed based on the applied AC signals to fully utilize the dynamic range of the ADC.

摘要: Site-specific modifications of proteins are desirable in biotechnological applications such as biopharmaceuticals, immunotherapy, vaccines, and are useful in chemical biology. Gluconoylation is a non-enzymatic, covalent, post-translational modification commonly observed on N-terminal His-Tags bearing proteins. We synthesized glucono-1,5-lactone derivatives, including azido variants for selective acylation. High yield acylation is achieved by simply mixing derivatives with target protein amidst diverse conditions of temperatures, aqueous buffers, excipients, or complex cell lysate.

摘要: An elution apparatus and a detection apparatus are described. The elution apparatus includes: a sample trap for trapping a sample; and one or more pumps and/or valves to move a liquid eluent and a liquid eluate, wherein the eluate includes an extracted portion of the sample that is extracted by the eluent. The detection apparatus includes: a capillary having a low-voltage (LV) end portion to receive a sample; and a conductivity detector coupled to a high-voltage (HV) end portion of the capillary to generate signals based on conductivity of a monitored portion of the capillary in the HV end portion, wherein the conductivity detector is electrically isolated from the LV end portion.
