This invention relates to an isolated nucleic acid fragment encoding an auxin transport protein. The invention also relates to the construction of a chimeric gene encoding all or a substantial portion of the auxin transport protein, in sense or antisense orientation, wherein expression of the chimeric gene results in production of altered levels of the auxin transport protein in a transformed host cell. The present invention also relates to methods using the auxin transport protein in modulating root development, and in discovering compounds with potential herbicidal activity.

The invention also provides methods of using the polynucleotides to modulate the level of WUS, improve transformation efficiency, to stimulate plant cell growth, including stem cells, to stimulate organogenesis, to stimulate somatic embryogenesis, to induce apomixis, and to provide a positive selection for cells comprising the polynucleotide. This invention relates generally to mechanisms of gene expression in plants and more specifically to regulation of expression of genes in plants in a “tissue-preferred” manner. A method for isolation of transcriptional regulatory elements that contribute to tissue-preferred gene expression is disclosed. Transcriptional regulatory elements isolated using the methods of this invention are demonstrated to direct tissue-preferred gene expression of genes within certain tissues of a plant. DNA molecules representing tissue-preferred transcriptional regulatory elements, vectors containing said DNA molecules and plants transformed with said vectors are demonstrated. Said transcriptional regulatory units are utilized to drive tissue-preferred expression of a gene that confers a selective advantage upon a plant.
