An imaging apparatus and method are provided. The probe for an imaging apparatus includes a manually manipulable proximal portion; a straight distal portion with a distal tip for locating at a site to define an observational field; and a curved portion between the proximal portion and the distal portion. The imaging method includes the steps of locating a distal tip of an imaging probe at a site to define an observational field; irradiating the observational field from the distal tip; and collecting a return signal at the distal tip; wherein the probe comprises a manually manipulable proximal portion. The apparatus and method provided herein are useful for various applications including but not limited to endomicroscopy and other microsurgical procedures performed under optical stereoscopic magnified visualization, such as neurosurgery, ENT/facial surgery and spinal surgery.

The ophthalmic surgical microscope (100) includes an OCT-system (140) for recording images of the object region (108). The OCT-system (140) includes an OCT-scanning beam (142) which is guided via a scan mirror arrangement (146) to the object region (108). An optic element (147) is provided between the scan mirror arrangement (146) and the microscope main objective (101). This optic element (147) bundles the OCT-scanning radiation exiting from the scan mirror arrangement (146) and transfers the same into a beam path which passes through the microscope main objective (101). Alternatively or in addition, the ophthalmic surgical microscope (100) includes an ophthalmoscopic magnifier lens (132) which can be pivoted into and out of the viewing beam path (105) and the OCT-scanning beam (142).

Functional tissue areas in a tissue region are located by illuminating the tissue by measurement illumination. Stimulation in the functional tissue areas leads to a change in at least one optical property of the reflected measurement illumination compared to the original measurement illumination. The functional tissue areas are located based on the change in the at least one optical property of the reflected measurement illumination by determining the difference between a stimulation image of the tissue region obtained during the stimulation and a comparison image of the tissue region obtained without stimulation. The difference may be formed from at least one image recorded with the measurement illumination and at least one image recorded without the measurement illumination to bring about an increase in contrast. An obtained image may be corrected on the basis of the determined topography of the tissue region to bring about an increase in contrast.
