摘要: Embodiments of the present disclosure are directed to apparatuses and methods for a pulsing system which can be configured to provide pressure pulses to a fluid communication tube to/from a bioreactor (for example). Such a system may comprise one or more of, and preferably a plurality of a pulsing motor, a compression plate, one or more fluid communications tubes comprising a biomedia fluid tube configured to direct biomedia fluid into and out of one or more bioreactors, and/or an intra-luminal fluid tube configured to direct intra-luminal fluid through a mandrel tube provided in each bioreactor, and one and/or another of a cam, gearing, and/or actuator configured to repetitively moving the compression plate so as to repetitively compress and release at least a portion of a respective intra-luminal fluid tube for a respective bioreactor to effect a temporary pressure increase (distension or strain) in a respective mandrel-tube according to the predetermined time period.

摘要: Provided is a bioreactor apparatus including: a liquid storage chamber for storing a liquid containing a first ion; a pump; a culture chamber for accommodating the liquid and cells to be cultured; and an ion-exchange chamber accommodating an ion-exchange substrate containing a second ion. Affinity of the second ion to the ion-exchange substrate is lower than affinity of the first ion to the ion-exchange substrate, or molar concentration of the second ion far outweigh molar concentration of the first ion. The storage chamber, the pump, the culture chamber and the ion-exchange chamber are connected via pipelines to form a closed loop, and the pump is configured to provide pressure to drive flow of the liquid in the closed loop. Also provided are a method for regulating ion concentration by the ion-exchange substrate and a method for cell culture by the bioreactor apparatus.

摘要: Cells may be grown in a bioreactor, and the cells may be activated by an activator (e.g., a soluble activator complex). Nutrient and gas exchange capabilities of a closed, automated cell expansion system may allow cells to be seeded at reduced cell seeding densities, for example. Parameters of the cell growth environment may be manipulated to load the cells into a particular position in the bioreactor for the efficient exchange of nutrients and gases. System parameters may be adjusted to shear any cell colonies that may form during the expansion phase. Metabolic concentrations may be controlled to improve cell growth and viability. Cell residence in the bioreactor may be controlled. In embodiments, the cells may include T cells. The cells may include T cell subpopulations, including regulatory T cells (Tregs), for example.

The system also includes a controller configured to operate the spinning membrane to receive biological fluid and to direct the target cells to the first container, to pause to permit magnetic particles to be associated with the target cells in the first container, to operate the spinning membrane to receive the contents of the first container with the magnet applied to the target cells associated with the magnetic particles, to remove or deactivate the magnet, and to transfer the target cells to the second container.

摘要: A plate cleaning device (1) for cleaning a clamp plate of a fermenter, wherein the clamp plate has a plurality of receptacles each configured to hold a bioreactor vessel, and at least one conduit with a conduit opening per receptacle. The device comprises a plate seat member (3), a cover member (2), a plurality of valve units (4), a first fluid terminal (71), a second fluid terminal (74) and a control unit (9). The plate seat member (3) is configured to hold the clamp plate (6) in a predefined position and orientation. The cover member (2) is configured to close the plate seat member (3) such that the clamp plate (6) is tightly enclosed by the plate seat member (3) and the cover member (2). The plurality of valve units (4) is configured to be coupled to the cover member (2) such that at least one valve unit (4) is associated to each of the conduit openings (62) of the clamp plate (6). The first fluid terminal (71) is configured to receive a first fluid container and the second fluid terminal (74) to receive a second fluid container. The cover member (2) is equipped with channels each having a first open end and a second open end. Each of the first open ends of the channels of the cover member (2) is positioned to be connected to one of the conduit openings (62) of the clamp plate (6). The valve units (4) are configured to be coupled to the second open ends of the channels of the plate such that all of the at least one conduit openings (62) per receptacle are associated to one valve (4). The first fluid terminal (71) and the second fluid terminal (74) are coupled to each of the valve units (4). The control unit (9) is configured to selectively connect the first fluid terminal (71) and the second fluid terminal (74) to the second open ends of the channels of the cover member (2).

The system is remarkable in that the cyclic gas pressure system comprises a feedthrough with a pipe configured to deviate the air influx from the one or more cells and/or organotypic cultures.

摘要: A device which automatically performs a step in which expanded and cultured cells are diluted to a desired cell concentration and re-inoculated using a cell-concentration adjustment device having an inlet for taking in a cell suspension; an outlet for discharging a diluted cell suspension; and a flow path which is provided between the inlet and the outlet and is capable of holding a cell suspension, the flow path being provided with: a liquid delivery pump for causing a cell suspension inside to flow; a cell-concentration measurement instrument for collecting data related to a cell concentration per unit amount of the cell suspension; and a dilution-liquid container for holding a dilution liquid which is supplied to the flow path to dilute the cell suspension. The device further includes a control unit for controlling at least the liquid delivery pump on the basis of the data obtained by the cell-concentration measurement instrument, wherein the control unit determines, on the basis of the data obtained by the cell-concentration measurement instrument, an amount of the dilution liquid required to bring the cell concentration to the desired concentration, and drives the liquid delivery pump so as to take in the required amount of the dilution liquid into the flow path and mix the cell suspension and the dilution liquid.
