Abstract: The invention relates to a process for preparing core-shell particles comprising the steps of (i) providing a dispersion of primary magnetic particles having a mean diameter lower than 200 nm in a solvent; (ii) adding one or more (semi-)metal (oxyhydr)oxide(s) and/or one or more precursor(s) of a (semi-)metal (oxyhydr)oxide to said dispersion; (iii) optionally adding a hydrolysis agent for said one or more precursor(s); (iv) injecting the dispersion in a spray dryer; whereby a (semi-)metal (oxyhydr)oxide shell is formed on the magnetic particles during spray drying. The invention also relates to particles obtainable by said process, to a formulation of said particles in a solvent and to the use of said particles or said formulation for RNA or DNA extraction.

Abstract: Provided are high throughput methods for physically linking cDNA molecules derived from mRNA molecules expressed by the same cell, and libraries of linked cDNA molecules produced by the methods. The methods comprise reverse transcribing mRNA from a single cell in a first container to produce cDNA molecules, and linking the cDNA molecules in a second container. The methods unexpectedly produced libraries of cDNA molecules with an increase in the number of molecules that are correctly linked to other molecules derived from the same cell.

Ferner umfasst das Verfahren ein Hybridisieren zumindest eines Teils der Target-Nukleinsäure (12) mit einer der Extraktionsnukleinsäuren (16) und Binden der Target-Nukleinsäure (12) über die Extraktionsnukleinsäure (16) an eines der magnetischen Mikropartikeln (10). Außerdem umfasst das Verfahren ein Bereitstellen eines Magnetfeldes in dem Reaktionsgefäß (22) derart, dass zumindest ein Teil der mit der Target-Nukleinsäure (12) verbundenen magnetischen Mikropartikeln (10) an einer Gefäßwand (22a, 22b, 22c) des Reaktionsgefäßes (22) sedimentiert.
