Abstract: The present disclosure provides immunogenic compositions and vaccines effective for reducing the incidence of or severity of clinical signs or symptoms of infection by a tick-borne pathogen selected from the group consisting of Rickettsia rickettsii, Ehrlichia chaffeensis, Ehrlichia canis, Ehrlichia ruminantium, Anaplasma marginale, and Anaplasma phagocytophilum. In preferred forms, the immunogenic composition comprises an inactivated whole cell bacteria selected from the group consisting of Rickettsia rickettsii, Ehrlichia chaffeensis, Ehrlichia canis, Ehrlichia ruminantium, Anaplasma marginale, Anaplasma phagocytophilum, and any combination thereof, together with an adjuvant.

Abstract: A cell-based assay for identifying a compound that inhibits iron transport in Gram-negative bacteria, engineered bacterial cells, and kits for conducting the same. The assay involves contacting a candidate compound with an engineered Gram-negative bacteria in the presence of iron for a sufficient period of time, exposing the reaction solution to an energy source to generate the detectable signal, and detecting changes in the detectable signal in the reaction solution over time. The engineered Gram-negative bacteria comprises an iron transport protein on its outer membrane that comprises an amino acid residue that has been engineered with a detectable label that generates a detectable signal. The changes in the detectable signal in the assay system over time correspond to the effect of the candidate compound on iron transport in the Gram-negative bacteria.

Abstract: A fluid-filled hollow optical fiber cell broadly includes a hollow-core optical fiber and a fluid. The optical fiber presents first and second fiber ends and a longitudinal passageway that extends continuously between the fiber ends. The fluid occupies the passageway, with the fiber ends being closed to hermetically seal the fluid within the optical fiber. The first fiber end has a rounded closed shape formed by at least partly melting the first fiber end to form melted fiber material, with the fiber material being permitted to solidify without splicing the first fiber end to another fiber so that the fiber material terminates the passageway at the first fiber end.

Abstract: The present invention is concerned with PCR-based detection methods and kits for the identification, differentiation, and quantification of different bacterial strains (e.g., Gram-negative bacterial strains), and also association of two or more PCR-positive genes to a single genome. The methods generally comprise carrying out PCR reactions using at least a first PCR primer set and/or probe for at least one target nucleic acid; and a second PCR primer set and/or probe for at least a second target nucleic acid. Positive PCR reaction products are then detected to determine test samples containing positive PCR reaction products for both the first and second target nucleic acids. This information can be used to calculate the gene association rate to determine whether the sample contains, for example, Shiga toxin-producing E. coli of the O-type serogroup.

Abstract: A new species of circovirus, porcine circovirus type 3 (PCV3), was identified from sows with clinical symptoms normally associated with porcine circovirus type 2 (PCV2) infection and in aborted fetuses. Molecular and serological analyses suggest PCV3 commonly circulates in U.S. swine. The present disclosure provides immunological compositions and methods related to the production and administration of such compositions.
